Validation of an in-house bovine serum enzyme immunoassay for progesterone measurement

Summary: The study aimed to develop and validate an in-house competitive enzyme immunoassay (EIA) for measuring circulating progesterone (P4) in dairy cattle serum, with a secondary objective of validating a commercial EIA. Results showed strong correlations between the in-house and commercial EIA with the gold standard radioimmunoassay (RIA), but weaker correlations at smaller concentrations. The in-house EIA demonstrated good accuracy for P4 measurement at concentrations above 0.4 ng/mL and perfect agreement with RIA using a threshold of 1.0 ng/mL.

Characterization of pregnancy-associated glycoproteins and progesterone as a predictor of twins and conceptus loss in high-risk-pregnancy Holstein cows

Summary: This study aimed to predict pregnancy loss and twins in Holstein cows with high-risk pregnancies by measuring plasma concentrations of PAG and progesterone. Results indicated that progesterone concentration at day 37 post-AI had moderate accuracy in predicting pregnancy loss and twins, while the variability in PAG concentrations was not sufficient for prediction in Holstein lactating cows.

Validation study of canine serum cortisol measurement with the Immulite 2000 Xpi cortisol immunoassay

Summary: The validation of canine serum cortisol determination using the Immulite 2000 Xpi cortisol immunoassay showed increased imprecision with decreasing serum cortisol concentration, highlighting the importance of considering analytical performance parameters in result interpretation. Quality control material had acceptable levels of imprecision and minimal bias, with QC rules heavily dependent on the desired allowable total error (TEa). Further investigation is needed to determine which TEa(QCM) can be guaranteed by simple QC rules.
