Lifetime risk of autosomal recessive neurodegeneration with brain iron accumulation (NBIA) disorders calculated from genetic databases

Summary: This study estimated the lifetime risks of different types of recessive NBIA disorders by evaluating the allele frequencies of pathogenic variants in genes associated with NBIA. The results showed that the lifetime risks of these disorders are significantly higher than previous population-based estimates, with the highest risks observed for disorders caused by variants in PLA2G6, PANK2, and COASY. These findings are important for understanding the disease burden, allocating resources, and targeting interventions for NBIA.
