Anthraquinones inhibit cytochromes P450 enzyme activity in silico and in vitro

Yitong Liu et al.

Summary: This study identified inhibitors of five major CYP isoforms among 22 anthraquinones using in silico and in vitro methods, with most of the anthraquinones showing potent inhibition of CYP1A2. Specifically, emodin, rhein, and aloe-emodin were found to inhibit multiple CYP activities in human liver microsomes, potentially leading to in vivo inhibition due to their high plasma concentrations. These results enable rapid identification of potential CYP inhibitors and prioritize future in-depth studies.

Inhibitory Effects of Schisandra Lignans on Cytochrome P450s and Uridine 5′-Diphospho-Glucuronosyl Transferases in Human Liver Microsomes

Hyung-Ju Seo et al.

Summary: The study found that lignans from Schisandra can inhibit the activity of certain cytochrome P450 enzymes, with some glutathione trapping reactive metabolites. This information may help predict potential drug interactions between Schisandra lignans and other drugs mainly metabolized by P450 enzymes.

Obtusifolin, an Anthraquinone Extracted from Senna obtusifolia (L.) H.S.Irwin & Barneby, Reduces Inflammation in a Mouse Osteoarthritis Model

Jiho Nam et al.

Summary: Obtusifolin, derived from Senna obtusifolia seeds, has anti-inflammatory properties that can reduce cartilage damage in OA by regulating MMPs and Cox2 expression. In addition, it modulates OA through the NF-kappa B signaling pathway, suggesting it as a potential symptom reliever for OA.

GNINA 1.0: molecular docking with deep learning

Andrew T. McNutt et al.

Summary: Molecular docking software Gnina 1.0, utilizing convolutional neural networks as scoring functions, outperforms AutoDock Vina in redocking and cross-docking tasks when binding pockets are explicitly defined. The ensemble of CNNs shows good generalization to unseen proteins and ligands, producing scores that correlate well with known binding poses. The 1.0 version of GNINA is available under an open source license for use as a molecular docking tool.

Comprehensive Investigation of Stereoselective Food Drug Interaction Potential of Resveratrol on Nine P450 and Six UGT Isoforms in Human Liver Microsomes

Seung-Bae Ji et al.

Summary: Resveratrol exhibited stereoselective inhibition on certain cytochrome P450s and uridine 5'-diphosphoglucuronosyl transferases, with different enantiomers showing varying levels of inhibition on different enzymes. Trans-resveratrol specifically demonstrated noncompetitive inhibition on CYP3A and UGT1A1 activities.
