Candidates for Repurposing as Anti-Virulence Agents Based on the Structural Profile Analysis of Microbial Collagenase Inhibitors

Georgiana Nitulescu et al.

Summary: Pharmacological inhibition of bacterial collagenases (BC) is an effective strategy to block bacterial virulence without inducing drug resistance. In this study, the chemical structures of Clostridium perfringens collagenase A (ColA) inhibitors were analyzed and various scoring methods were used to predict new compounds with ColA inhibitory activity. The results showed that compounds containing at least one aromatic ring, particularly a nitrobenzene fragment, exhibit higher potency as BC inhibitors. Furthermore, molecular descriptors were identified as filters to identify potential BC inhibitors, and the DrugBank database was screened to identify 100 potential BC inhibitors, including 10 approved drugs. Benzthiazide, entacapone, and lodoxamide were selected as the most promising candidates for in vitro testing based on their pharmacotoxicological profiles.

A new era for the design of TRPV1 antagonists and agonists with the use of structural information and molecular docking of capsaicin-like compounds

Julio Caballero

Summary: The design of TRPV1 antagonists and agonists has advanced with the availability of near-atomic resolution TRPV1 structures. Understanding the ligand-binding forms of these compounds allows for a more thorough use of molecular modelling methods in rational drug design protocols. This study highlights the important interactions between TRPV1 and capsaicin-like compounds based on known pharmacophore and TRPV1 structures, offering valuable insights for the design of novel TRPV1 antagonists and agonists.

Melatonin and Selenium Suppress Docetaxel-Induced TRPV1 Activation, Neuropathic Pain and Oxidative Neurotoxicity in Mice

Kemal Ertilav et al.

Summary: Docetaxel has positive therapeutic actions in treating glioblastoma, breast tumors, and prostate cancers, but can induce neuropathic pain and neurotoxicity. TRPV1 cation channel activation mediates DT-induced pain, and melatonin and selenium protect against oxidative neurotoxicity and pain. Antioxidant properties of MT and Se play a key role in mediating these protective effects.

Dual regulation of TRPV1 channels by phosphatidylinositol via functionally distinct binding sites

Aysenur Torun Yazici et al.

Summary: Regulation of the TRPV1 channel by phosphoinositides is complex, with PtdIns potentially acting as a competitive antagonist or activator depending on the presence of PtdIns(4,5)P-2. Different binding sites for phosphoinositides may explain the varied effects observed on TRPV1 activity.

Summary: This study utilized cryo-EM to visualize conformational transitions of TRPV1, revealing the dynamic transitions of its selectivity filter in response to algogenic agents. The findings also demonstrate a general framework for understanding how TRP channels function as polymodal signal integrators.

Effects of Venlafaxine, Risperidone and Febuxostat on Cuprizone-Induced Demyelination, Behavioral Deficits and Oxidative Stress

Dragos Paul Mihai et al.

Summary: MS is a demyelinating, autoimmune disease affecting a large number of young adults, in need of novel therapies. Venlafaxine and risperidone were found to improve motor performance and sensitivity to a cold stimulus, while all drugs alleviated cuprizone-induced deficits in superoxide dismutase activity. Agonistic activity of febuxostat may be useful for discovering novel TRPA1 ligands.

TRPV1-Targeted Drugs in Development for Human Pain Conditions

Mircea Iftinca et al.

Summary: TRPV1 is a significant channel in pain perception and its agonists are being studied for chronic pain management. While TRPV1 antagonists have limited clinical use due to side effects, capsaicin offers a potential avenue for long-lasting pain relief by defunctionalising nociceptors. The development of TRPV1-targeted drugs could provide a new generation of analgesics for various pain conditions.
