The role of ROS in tumour development and progression

Eric C. Cheung et al.

Summary: Eukaryotic cells have complex systems to regulate reactive oxygen species (ROS), which are deregulated in cancer cells. ROS also modulate the tumor environment and affect stromal cells supporting metabolism, blood supply, and immune responses. The response to ROS is complex and depends on multiple factors, including ROS characteristics and tumor characteristics. Understanding the complexity of ROS in cancers is key to developing ROS-targeting therapies for cancer treatment.

Antitumor and toxicity study of mitochondria-targeted triptolide derivatives using triphenylphosphine (TPP+) as a carrier

Huina Song et al.

Summary: By using delocalized lipophilic cations as carriers, mitochondria-targeted derivatives of TP were designed and synthesized, maintaining strong antitumor activity with tumor cell selectivity. These derivatives also increased the release of lactate dehydrogenase and production of ROS, reduced mitochondrial membrane potential, and arrested HepG2 cells in the G0/G1 phase.

Engineered Cell-Penetrating Peptides for Mitochondrion-Targeted Drug Delivery in Cancer Therapy

Qicai Xiao et al.

Summary: Mitochondrion is a promising target for cancer therapy, and cell-penetrating peptides with mitochondrion-targeting ability are efficient tools for delivering therapeutics. New mitochondrion-targeting peptides (MTPs) have been synthesized, with MTP3 showing excellent mitochondrion-targeting ability and potential for drug delivery. The study validated the feasibility of MTP3 by preparing a mitochondrion-targeting prodrug that showed good cytotoxicity against tumor cells while having minimal toxicity towards normal cells.

The Regulation of ROS- and BECN1-Mediated Autophagy by Human Telomerase Reverse Transcriptase in Glioblastoma

Xuelu Ding et al.

Summary: Research indicates that hTERT plays a crucial role in glioblastoma progression by regulating the autophagy pathway, as downregulation of hTERT affects cellular autophagy levels and leads to cell death, while overexpression of BECN1 or the use of antioxidants can restore cell survival.
