Nrf2/Keap1/ARE signaling: Towards specific regulation

Summary: The Nrf2 transcription factor regulates the expression of genes involved in cell defense against oxidative stress, which is associated with various diseases. The interactions between Nrf2 and Keap1 protein play a crucial role in this regulation. The activation of the Nrf2 system is considered as a promising strategy to treat diseases related to oxidative stress, including COVID-19. Two approaches are being developed to inactivate the Keap1 guardian function and enhance Nrf2 activity.

Understanding the signal amplification in this pathway can help identify potential targets for preventing and treating oxidative stress-related diseases and facilitate health risk assessment of oxidative chemicals.

Delivery of Antibody-Like Molecules, Monobodies, Capable of Binding with SARS-CoV-2 Virus Nucleocapsid Protein, into Target Cells

Summary: Based on previous studies, two high-affinity monobody molecules capable of interacting with SARS-CoV-2 nucleocapsid protein were selected. Genetically engineered constructs containing monobody and TAT peptide were produced and expressed in E. coli. Through thermophoresis and cellular thermal shift assays, the construct with the highest affinity to the nucleocapsid protein was identified. Replacement of TAT peptide with S10 shuttle peptide significantly improved the construct's penetration into target cells.
