Machine learning models to predict sweetness of molecules

Mansi Goel et al.

Summary: Predicting the sweetness of small molecules is becoming increasingly important due to the rise of type-2 diabetes. We developed regression-based machine learning and deep learning algorithms and curated a dataset of 671 sweet molecules with known sweetness values. Our models using Gradient Boost and Random Forest Regressors achieved high accuracy with correlation coefficients of 0.94 and 0.92 respectively.

Predictive identification of co-formers in co-amorphous systems

Luke I. Chambers et al.

Summary: This study aimed to understand the properties of co-formers in co-amorphous pharmaceutical materials and predict co-amorphous system formation using a PLS-DA analysis based on 36 variables. The analysis revealed key variables that favor the formation of co-amorphous systems. Testing the predictive model with 29 potential co-formers showed a high accuracy rate of 90% in predicting co-amorphous nature.

Co-Amorphous Formulations of Furosemide with Arginine and P-Glycoprotein Inhibitor Drugs

Marika Ruponen et al.

Summary: In this study, ARG and P-gp inhibitors (VER, PIP, QRT) were used as co-formers for co-amorphous mixtures of the BCS class IV drug FUR. The solid-state properties and stability of the mixtures were characterized, showing that only the co-amorphous FUR:ARG mixtures were able to enhance both dissolution and permeation of FUR. This suggests that formulating co-amorphous salts with ARG may be a promising option for improving the solubility/permeability of poorly soluble/permeable FUR.

Preparation and Evaluation of Co-amorphous Formulations of Telmisartan-Amino Acids as a Potential Method for Solubility and Dissolution Enhancement

Mai Khanfar et al.

Summary: This study aimed to enhance the solubility and dissolution rate of Telmisartan (TLM) through the co-amorphous drug amino acid (AA) approach, with the TLM-arginine (ARG) formulation showing the greatest enhancement. The results indicated that the 1:2 w/w ratio of TLM-ARG co-amorphous had the best solubility and dissolution rate, while the 1:0.5 w/w ratio showed the best stability.

Considerations for the selection of co-formers in the preparation of co-amorphous formulations

Dani Lakshman Yarlagadda et al.

Summary: Co-amorphous drug delivery systems, utilizing small molecular weight compounds called co-formers to stabilize drugs in amorphous form, have shown potential as an alternative to amorphous solid dispersions technology. The stability and supersaturation potential of co-amorphous phases largely depend on the type of co-former employed, but the rationality behind co-former selection is poorly understood. Various factors affecting co-former selection in co-amorphous systems have been compiled, including thermodynamic, kinetic, and pharmacokinetic-pharmacologically relevant parameters.
