TRAF3 alterations are frequent in del-3′IGH chronic lymphocytic leukemia patients and define a specific subgroup with adverse clinical features

Claudia Perez-Carretero et al.

Summary: Interstitial 14q32 deletions involving IGH gene are rare events in CLL and are associated with poor prognosis. Multiple gene mutations were found in del-3 ' IGH CLLs, including NOTCH1, ATM, genes involved in the RAS signaling pathway, and TRAF3. Biallelic inactivation of TRAF3 was an independent risk factor for poor outcome in these patients.

The genomic landscape of canine diffuse large B-cell lymphoma identifies distinct subtypes with clinical and therapeutic implications

Diana Giannuzzi et al.

Summary: Giannuzzi et al. conducted an integrated analysis of clinical features and exome and RNA sequencing data in dogs with diffuse large B-cell lymphoma (DLBCL) to define the genetic landscape and identify mutations associated with outcome. The study found common signaling pathways and cellular processes with human DLBCL, but different frequencies of recurrently mutated genes. A prognostic model integrating genetic variants and clinical and transcriptomic features was developed to predict outcome in dogs with DLBCL. These findings provide a comprehensive understanding of the genetic drivers of canine DLBCL and potential therapeutic opportunities.

Summary: This study identifies a syndrome of immune dysregulation that is associated with recurrent bacterial infections, autoimmunity, systemic inflammation, B cell lymphoproliferation, and high levels of immunoglobulin. The mutations in the TRAF3 gene and common genetic variants associated with reduced TRAF3 expression both contribute to the development of this syndrome. These findings shed light on the role of TRAF3 in human immunity and its impact on various diseases.

TNF Receptor Associated Factor 2 (TRAF2) Signaling in Cancer

Daniela Siegmund et al.

Summary: TRAF2 is a protein that interacts with TNFR2 and other TNFRSF receptors, and is involved in the activation of multiple signaling pathways. It plays a role in NFkB pathway, MAP kinase cascades, ER stress signaling, autophagy regulation, and cell death control. TRAF2 can function as a scaffold and E3 ligase, and is implicated in the oncogenesis and tumor suppression of various cancers. Targeting TRAF2 shows potential for therapeutic purposes.
