Th1-involved immune infiltrates improve neoadjuvant chemoradiotherapy response of esophageal squamous cell carcinoma

Jianye Yuan et al.

Summary: This study found that Th1-cell infiltration is associated with pathological complete response (pCR) in esophageal squamous cell carcinoma (ESCC) patients, and Th1 cells regulate the chemosensitivity and radiosensitivity of ESCC cells. The study also identified active and desert subtypes based on immune cell infiltration, which had different pCR rates. These findings suggest that increasing Th1-cell infiltration may improve the response to neoadjuvant chemoradiotherapy (NCRT).

Ferroptosis-Related lncRNA Signature Correlates with the Prognosis, Tumor Microenvironment, and Therapeutic Sensitivity of Esophageal Squamous Cell Carcinoma

Jiahao Zhu et al.

Summary: Esophageal squamous cell carcinoma (ESCC) is closely associated with poor survival, and ferroptosis, an iron-dependent mode of cell death, plays an important role in the biological behavior of ESCC cells. This study aimed to explore the clinical significance of ferroptosis-related long noncoding RNAs (FRLs) in ESCC. Through RNA sequencing and coexpression analysis, a ten-FRL prediction signature was established and validated using multiple databases. The risk model showed promise as a predictor of prognosis and immunotherapeutic response in patients with ESCC.

Characterization of the Intra-tumoral B Cell Immunoglobulin Repertoire Is of Prognostic Value for Esophageal Squamous Cell Carcinoma

Zhizhong Wang et al.

Summary: This study explored the repertoire of tumor infiltrating B cells (TIBs) in esophageal squamous cell carcinomas (ESCC) and identified several indicators associated with prognosis. Higher levels of immunoglobulin expression, CD138 expression, IgG2-producing plasma cells, CSR events, and clone diversity were all associated with better survival. CD20-negative IgG2-producing plasma cells had a strong positive effect on overall survival.

Irradiation induces DJ-1 secretion from esophageal squamous cell carcinoma cells to accelerate metastasis of bystander cells via a TGF-β1 positive feedback loop

Junjie Gu et al.

Summary: DJ-1 is highly expressed in the serum and cell supernatants of patients with ESCC, and promotes ESCC metastasis through the DJ-1/HSC70/Smad3 signaling axis. The TSP1/TGF-beta 1/Smad3 positive feedback pathway is the core pathway that promotes ESCC metastasis. DJ-1 can serve as a potential therapeutic target for reversing RIBE and predicting the efficacy of radiotherapy.

A tumor immune microenvironment-related integrated signature can predict the pathological response and prognosis of esophageal squamous cell carcinoma following neoadjuvant chemoradiotherapy: A multicenter study in China

Peng Wu et al.

Summary: Our study identified a promising three-gene-based predictive signature that has powerful clinical implications for the identification of pCR and a good prognosis among patients with ESCC. Patients with a high signature score had lower PD-L1 and IDO1 expression levels but higher CD8(+) T cells infiltration, suggesting a negative correlation between PD-L1 and IDO1 with a high signature score and their associations with pCR and a better prognosis.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

High SAA1 Expression Predicts Advanced Tumors in Renal Cancer

Sen Li et al.

Summary: Renal cell carcinoma (RCC) is the most frequent malignant tumor of the kidney, with 30% of patients being diagnosed at an advanced stage. Clear cell renal cell carcinoma (ccRCC) is the most common subtype, and SAA1 has been identified as a potential diagnostic and prognostic marker for advanced ccRCC. Studies show that high levels of SAA1 are associated with poor overall and disease-free survival times in advanced ccRCC patients. Targeting SAA1 may offer a novel therapeutic approach for these patients.

NETO2 promotes esophageal cancer progression by inducing proliferation and metastasis via PI3K/AKT and ERK pathway

Jia-cheng Xu et al.

Summary: Our study revealed that high expression of NETO2 in ESCC patients was associated with tumor staging and lymph node metastasis, promoting ESCC progression through enhancing cell proliferation, suppressing apoptosis, and stimulating tumor growth. Additionally, NETO2 was found to play a crucial role in regulating migration, invasion, and epithelial-mesenchymal transition in ESCC cells. Mechanistically, NETO2 activated ERK, PI3k/AKT, and Nrf2 signaling pathways, with Nrf2 identified as a critical downstream mediator responsible for tumorigenesis. This suggests that NETO2 may serve as a potential therapeutic target or prognostic biomarker for ESCC patients.
