Summary: Primary central nervous system-diffuse large B-cell lymphoma (PCNS-DLBCL) is a rare malignant lymphoma with a poor prognosis. The tumor microenvironment (TME) and the PD-1/PD-L1 immune checkpoint pathway have significant impacts on the prognosis of PCNS-DLBCL, with increased CD4+ T cells and PD-L1-expressing immune cells associated with better outcomes. Further studies with larger sample sizes are needed to evaluate the potential role of targeted therapy and immune checkpoint inhibitors in this disease.

Summary: Tumor-associated macrophages (TAMs) are associated with poor prognosis in diffuse large B-cell lymphoma (DLBCL). Different infiltration patterns of macrophages in the tumor microenvironment are correlated with survival outcomes, with high M2c-like macrophage infiltration and low FOXP3-positive Treg infiltration linked to a worse prognosis. Additionally, PTX3 is identified as a new prognostic biomarker in DLBCL.

Summary: This study investigated the clinicopathological risk factors for a poor prognosis in elderly patients with malignant lymphoma of the central nervous system. The results showed that Karnofsky Performance Status less than 60% 3 months after treatment, double expressor lymphoma, expression of programmed death-ligand 1 in tumor infiltrating lymphocytes or tumor-associated macrophages, and Epstein-Barr virus infection were important risk factors for poor prognosis.

Summary: This study investigated the role of Liver-X-Receptors (LXRs) in diffuse large B-cell lymphomas (DLBCL) and found an association of the NR1H3 gene with the tumor microenvironment. Analysis of large DLBCL datasets revealed that patients with high expression of NR1H3 had longer survival. Additionally, analysis of macrophage gene expression profiles showed significant transcriptional differences between subgroups.

Clinical and Biological Subtypes of B-cell Lymphoma Revealed by Microenvironmental Signatures
