Methionine Restriction: Ready for Prime Time in the Cancer Clinic?

Summary: Efforts to selectively starve cancer cells have been made for a century. Calorie-restriction or low-carbohydrate diets have limited success. Methionine restriction, especially in combination with chemotherapy, has shown promising results in preclinical studies. The question remains whether methionine restriction, either through diet or with the help of methioninase, is ready for clinical use, particularly for advanced cancer patients.

Methionine restriction enhances the chemotherapeutic sensitivity of colorectal cancer stem cells by miR-320d/c-Myc axis

Summary: This study found that methionine restriction (MR) combined with 5-fluorouracil (5-FU) chemotherapy can enhance the chemotherapeutic sensitivity of colorectal cancer stem cells (CRC-SCs). The up-regulation of miR-320d and inhibition of c-Myc expression were identified as the molecular mechanisms underlying this effect. These findings provide a molecular basis for the regulation of chemotherapeutic sensitivity of CRC-SCs by MR.

A gene signature is critical for intrahepatic cholangiocarcinoma stem cell self-renewal and chemotherapeutic response

Summary: This study identified four ICC stemness-associated genes (SDHAF2, MRPS34, MRPL11, and COX8A) that are significantly upregulated in ICC tissues and negatively associated with clinical outcome. Functional studies indicated that these genes are associated with self-renewal ability of ICC and transgenic expression could enhance chemoresistance. Mechanistically, the genes' pro-stemness effect requires the activation of the methionine cycle and is dependent on MAT2A. A novel nomogram was established to evaluate the effectiveness of TACE for ICC patients. Dietary methionine intervention studies suggested that patients with high nomogram score might benefit from methionine restriction.
