Cell-Cell Interaction-Mediated Signaling in the Testis Induces Reproductive Dysfunction-Lesson from the Toxicant/Pharmaceutical Models

Lingling Wang et al.

Summary: Emerging evidence suggests that cell-cell interactions, mediated by signaling proteins including FAK, Akt1/2, and Cdc42 GTPase, play a crucial role in supporting spermatogenesis. Manipulation of these signaling proteins can modify the outcome of spermatogenesis, offering potential therapeutic approaches for toxicant-induced male reproductive dysfunction.

Basal Autophagy Is Necessary for A Pharmacologic PPARα Transactivation

Eun Young Kim et al.

Summary: This study reveals the impact of autophagy defects on the transactivation of PPAR alpha, which plays a key role in fasting-associated metabolisms. Autophagy defects lead to the activation of NRF2, an antioxidant transcription factor, which in turn blunts the function of PPAR alpha. These findings highlight the importance of basal autophagy for the function of PPAR alpha.

Gap junction intercellular communication mediates cadmium-induced apoptosis in hepatocytes via the Fas/FasL pathway

Fan Yu et al.

Summary: This study found that gap junction inhibitor may play a dual role in Cd-induced cell damage by inhibiting the transmission of death signals from damaged cells to healthy cells but also aggravating the transmission of death signals between damaged cells. The Fas/FasL-mediated death receptor pathway may play an important role in this process.

Cadmium as a testicular toxicant: A Review

Jitender Kumar Bhardwaj et al.

Summary: Cadmium is a toxic heavy metal that accumulates in organs such as the liver, kidney, and testes, leading to perturbed functioning. In the testes, cadmium induces oxidative stress, apoptosis of spermatogenic cells, reduction in androgen production, and impaired sperm functions. Understanding how cadmium toxicity affects various testicular processes is crucial for developing preventative measures to promote fertility among males.

Zearalenone Exposure Disrupts Blood-Testis Barrier Integrity through Excessive Ca2+-Mediated Autophagy

Jinjin She et al.

Summary: The study demonstrated that exposure to Zearalenone (ZEA) resulted in severe testicular damage and a decrease in the expression of blood-testis barrier (BTB) junction-related proteins in a time-dependent manner, accompanied by an increase in autophagy-related proteins. In vitro, ZEA treatment led to an increase in cytoplasmic Ca2+ levels, autophagy markers LC3-II and p62 levels, and a decrease in BTB junction proteins, with the dislocation of the gap junction protein Cx43. Inhibition of autophagy or cytoplasmic Ca2+ was able to mitigate the effects of ZEA on BTB in TM4 cells.

Summary: This study demonstrates that in rat cerebral cortical neurons, the mechanisms underlying Cd-induced Fas-mediated activation of the mitochondrial apoptotic pathway involve the activation of caspase-8 and the c-Jun N-terminal kinase (JNK) pathway.
