The inhibitor of κB kinase β (IKKβ) phosphorylates IκBα twice in a single binding event through a sequential mechanism

Summary: Phosphorylation of IκB proteins by IKKβ leads to IκB degradation and activation of nuclear factor κB transcription factors. The phosphorylation of IκBα at Ser32 and Ser36 by IKKβ was investigated. The kinetic analysis showed a fast exponential phase attributed to phosphorylation at Ser32 and Ser36, and a slower linear phase attributed to phosphorylation at other residues. Phosphorylation of Ser32 increased the phosphorylation rate of Ser36.

Global change in hepatitis C virus prevalence and cascade of care between 2015 and 2020: a modelling study

Summary: This study aims to evaluate the burden of hepatitis C virus (HCV) in 2020 and forecast the burden by 2030. Using literature review, Delphi process, and mathematical modelling, it estimated the global prevalence of viraemic HCV infection to be 0.7% in 2020, corresponding to 56.8 million infections. The findings suggest that the current efforts are not on track to achieve the global elimination targets by 2030.

Interlocking activities of DNA polymerase β in the base excision repair pathway

Summary: Base excision repair (BER) is a crucial cellular pathway for DNA damage repair, and DNA polymerase beta (Pol beta) plays a key role in this process. This study reveals the sequential order of two enzymatic activities of Pol beta during BER and identifies the intermediate molecules involved. The coordinated activities of Pol beta's active sites are found to be critical for the BER pathway, suggesting potential alternative strategies for therapeutic intervention.
