A Novel 16-Genes Signature Scoring System as Prognostic Model to Evaluate Survival Risk in Patients with Glioblastoma

Zunpeng Yu et al.

Summary: This study aimed to identify prognostic survival-related genes for glioblastoma (GBM) and construct a survival risk prediction model. By analyzing gene expression data and clinical information, 99 genes associated with overall survival were identified and a model with 16 genes was established. The model showed superior prediction ability compared to existing signatures and the genes could potentially serve as transcriptional biomarkers, aiding in personalized treatment for GBM patients.

Class I HDAC overexpression promotes temozolomide resistance in glioma cells by regulating RAD18 expression

Daniela Hanisch et al.

Summary: This study reports a novel mechanism by which overexpression of class I HDACs increases resistance of glioblastoma cells to TMZ. HDACs promote the bypass of O-6-methylguanine DNA lesions by enhancing RAD18 expression, leading to resistance to TMZ treatment.

Prognostic Neurotransmitter Receptors Genes Are Associated with Immune Response, Inflammation and Cancer Hallmarks in Brain Tumors

Yuri Belotti et al.

Summary: This study investigates the role of dysregulated neurotransmitter receptors (NTRs) genes in the prognosis of low-grade glioma (LGG) and glioblastoma multiforme (GBM). The findings suggest that certain NTR genes are associated with different patient survival outcomes in LGG and GBM. These genes show downregulation across glioma grades and have a negative correlation with genes related to immune response, inflammation, and cancer hallmarks in LGG. The study highlights the potential implications for therapeutic and preventive strategies targeting the regulatory networks involving the autonomic nervous system, cancer cells, and the tumor microenvironment.

Identification of Core Genes and Screening of Potential Targets in Glioblastoma Multiforme by Integrated Bioinformatic Analysis

Ji'an Yang et al.

Summary: The study identified differentially expressed genes related to neural cell signal transduction through reanalysis of GBM and LGG datasets, and highlighted the significance of core molecules in ionic signaling pathways. The research further validated the significant relationship between these core genes and patient survival, and identified drugs related to cancer and neuropsychiatric diseases through database analysis. Further exploration of these core genes is needed to understand their potential in diagnosis, prognosis, and targeted therapy for neurological tumors and related diseases.

Potential Diagnostic Value of the Differential Expression of Histone H3 Variants between Low- and High-Grade Gliomas

Irati Hervas-Corpion et al.

Summary: The study identified that high expression levels of H3.1/H3.2 are associated with glioblastomas, while high expression of H3.3 is linked to low-grade gliomas. There is a potential for further investigation on the association between H3 variants and glioma aggressiveness. Additionally, the reduced levels of H3.3 in glioblastomas compared to other gliomas suggest a potential diagnostic utility in distinguishing high-grade gliomas.

Analysis of intraoperative human brain tissue transcriptome reveals putative risk genes and altered molecular pathways in glioma-related seizures

Anteneh M. Feyissa et al.

Summary: In this study, altered processes such as cell-to-cell signaling, inflammation-related, and glutamatergic neurotransmission were identified in the pathogenesis of GRS. DEGs specific to GRS included overexpression of genes involved in cell-to-cell and glutamatergic signaling, and down-regulation of genes involved in immune-trafficking. Target genes identified offer new landscape for further study in the fields of brain tumors and seizures.

The Impact of Epigenetic Modifications on Adaptive Resistance Evolution in Glioblastoma

Qiong Wu et al.

Summary: Glioblastoma is highly resistant to standard therapies and the durability of response to the best chemotherapy agent, Temozolomide, is often short-lived due to tumor resistance. The need for therapies that can provide synergy to chemoradiation is urgent, as adaptive resistance evolution in GBM is facilitated through treatment-induced epigenetic modifications. Understanding and targeting these epigenetic modifications associated with GBM resistance is a top priority.

The role of epigenetic modifications, long-range contacts, enhancers and topologically associating domains in the regulation of glioma grade-specific genes

Ilona E. Grabowicz et al.

Summary: Genome-wide studies have revealed specific genetic alterations, transcriptomic patterns, and epigenetic profiles associated with different types of glioma. The organization of chromatin and epigenetic landscape of enhancers have been shown to impact gene expression in low grade versus high grade gliomas. The identification of transcription factors and genes regulated by long-range contacts with enhancers provides important insights into the epigenetic differences associated with differential gene expression in gliomas of different malignancy.
