Summary: The microenvironment of malignant melanomas plays a crucial role in the infiltration and vascular dissemination of immune and cancer cells. In this study, we found that the complement system is activated in melanoma patients and murine melanomas. Complement-derived C5a recruits neutrophils, and activated neutrophils release neutrophil extracellular traps (NETs), which open the endothelial barrier and facilitate the entrance of melanoma cells into circulation. Inhibition of neutrophil activation could prevent vascular intravasation of melanoma cells.

Melanoma Stem Cells Educate Neutrophils to Support Cancer Progression

Summary: In melanoma patients, poor prognosis often correlates with high presence of cancer-associated neutrophils, indicating that tumors can recruit these immune cells to specifically sustain their own development and progression. However, the role of cancer stem cells (CSCs) in this dialogue has not been elucidated yet. Our results revealed that melanoma SCs can reshape the immune microenvironment by triggering a pro-tumor N2 phenotype in neutrophils, which in turn are able to confer stemness properties to melanoma cells.

Levels of colonic formyl peptide receptor 1 and annexin A1 are correlated with histological recovery in Crohn's disease patients under remission. Infliximab may induce annexin A1 expression and secretion in activated intestinal leukocytes, potentially stimulating epithelial formyl peptide receptors and promoting wound healing in inflammatory bowel disease patients.
