Transcriptome-wide subtyping of pediatric and adult T cell acute lymphoblastic leukemia in an international study of 707 cases

Yu-Ting Dai et al.

Summary: This study provides insights into the molecular landscape of T cell acute lymphoblastic leukemia (T-ALL) by analyzing the transcriptomic profiles of 707 patients. The researchers identified 10 subtypes of T-ALL and associated them with different stages of T cell development. They also found that adult patients had more cooperative mutations in epigenetic regulators and genes involved in JAK-STAT and RAS signaling pathways, suggesting a mixed phenotype acute leukemia nature.

Monoallelic Heb/Tcf12 Deletion Reduces the Requirement for NOTCH1 Hyperactivation in T-Cell Acute Lymphoblastic Leukemia

Diogo F. T. Veiga et al.

Summary: Early T-cell development is controlled by E proteins, NOTCH1 and pre-TCR signaling. Perturbations of these pathways are implicated in leukemogenesis. This study investigates the individual roles of HEB, E2A, NOTCH1, and Tcf12 in T-ALL using gene-deficient mice. The findings suggest that HEB and E2A restrain cell proliferation, Tcf12 gene dosage is essential for the clonal expansion of pre-leukemic stem cells, and HEB acts as a tumor suppressor in T-ALL.

A comprehensive analysis of LMO2 pathogenic regulatory profile during T-lineage development and leukemic transformation

Wenhao Wang et al.

Summary: This study systematically investigated the performance of LMO2 during T-lineage development in the pre-leukemic stage. The data indicated that LMO2 caused transcriptional dysregulation primarily in DN3 thymocytes, and interacted with RUNX1 and ASH2L proteins. However, the binding of LMO2 and its interactions gradually decreased in the following stages. Moreover, LMO2 showed different effects on cellular behavior in different stages.

Thymic Epithelial Cell-Derived IL-15 and IL-15 Receptor α Chain Foster Local Environment for Type 1 Innate Like T Cell Development

Huishan Tao et al.

Summary: Expression of cytokines and receptors in thymic epithelial cells may impact T cell development. IL-15 and IL-15Rα play a crucial role in the development of type 1 innate-like T cells, and TECs may influence thymic inflammatory innate-like T cells by trans-presenting IL-15.

IL-15Rα-Independent IL-15 Signaling in Non-NK Cell-Derived IFNγ Driven Control of Listeria monocytogenes

Madhuparna Nandi et al.

Summary: Interleukin-15 (IL-15) regulates immune cell activity by forming a trimeric complex with IL-15Rα, but can also signal through the IL-15Rβγ(c) dimer. IL-15 deficiency results in the inability to control bacterial infections, while mice lacking IL-15Rα are able to effectively clear pathogens.

Summary: Combination therapy with ruxolitinib and venetoclax shows promise for treating T-ALL, but additional inhibition of the CXCR4-CXCL12 axis may be needed to maximize the possibility of complete remission.
