Hyperglycaemia induces metabolic reprogramming into a glycolytic phenotype and promotes epithelial-mesenchymal transitions via YAP/TAZ-Hedgehog signalling axis in pancreatic cancer

Summary: Hyperglycaemia in pancreatic ductal adenocarcinoma (PDAC) patients can induce metabolic reprogramming, leading to epithelial-mesenchymal transition (EMT) and chemoresistance. This process involves the overexpression of yes-associated protein (YAP)/PDZ-binding motif (TAZ) and activation of the Hedgehog pathway. Additionally, hyperglycaemia stimulates the production of collagen fibre-related proteins in cancer-associated fibroblasts through YAP/TAZ expression. Inhibiting YAP/TAZ can reverse EMT, chemoresistance, and create a less favorable tumor microenvironment under hyperglycemic conditions.
