The combination of PD-1 blockade with interferon-α has a synergistic effect on hepatocellular carcinoma

Summary: Combining Peg-IFN alpha with PD-1 blockade enhances T-cell infiltration, improves the efficacy of PD-1 antibody, and prolongs survival in HCC mouse models. Peg-IFN alpha recruits CD8(+) T cells to the HCC microenvironment by inducing tumor cells to secrete the chemokine CCL4. However, the HCC microenvironment overcomes the immune response by upregulating PD-1 expression in CD8(+) T cells. The combination of PD-1 blockade with Peg-IFN alpha restores the cytotoxic capacity of CD8(+) T cells and exhibits a significant synergistic effect on HCC.

These insights also shed light on immunotherapy-mediated immunoediting and the dual opposing roles of IFNs in immune checkpoint blockade response.
