Maxillo-Facial Morphology in Beckwith-Wiedemann Syndrome: A Preliminary Study on (epi)Genotype-Phenotype Association in Caucasians

Patrizia Defabianis et al.

Summary: This study aimed to investigate the association between maxillo-facial morphology and growth pattern with genotype in BWS and macroglossia patients. The results showed that patients with IC2-LoM and a negative genotype had a more marked tongue enlargement, while UPD(11)pat patients showed mild macroglossia. Cluster analysis revealed different clusters of BWS patients based on cephalometric characteristics. Children with IC2-LoM or a negative genotype displayed a negative prognostic score.

Prevalence of (Epi)genetic Predisposing Factors in a 5-Year Unselected National Wilms Tumor Cohort: A Comprehensive Clinical and Genomic Characterization

Janna A. Hol et al.

Summary: This study found that (epi)genetic predisposing factors were present in at least 33.3% of pediatric patients with Wilms tumor in the Netherlands. Multiple predisposing genes associated with WT were identified, but further validation is needed.

Clinical and molecular characterization of patients affected by Beckwith-Wiedemann spectrum conceived through assisted reproduction techniques

Diana Carli et al.

Summary: The prevalence of Beckwith-Wiedemann spectrum (BWSp) is higher in children conceived through assisted reproductive techniques (ART), and these ART-conceived BWSp patients may have a milder phenotype. Furthermore, the prevalence of BWSp has increased over time, potentially due to the increase in ART usage.

Molecular Basis of Beckwith-Wiedemann Syndrome Spectrum with Associated Tumors and Consequences for Clinical Practice

Thomas Eggermann et al.

Summary: Beckwith-Wiedemann syndrome is a congenital overgrowth disorder caused by molecular alterations in chromosome 11p15.5. The molecular subtype of the syndrome determines the phenotype and tumor risk. Molecular diagnosis is challenging due to its limitations and challenges.

Improved molecular detection of mosaicism in Beckwith-Wiedemann Syndrome

Samuel W. Baker et al.

Summary: Beckwith-Wiedemann Syndrome (BWS) is characterized by overgrowth and tumor predisposition, mainly caused by methylation defects in imprinting control regions on chromosome 11p15.5. Molecular diagnosis of BWS requires the use of multiple sensitive diagnostic techniques to reliably detect low-level aberrations. Testing both blood and solid tissue samples can increase the diagnostic yield and identify mosaic disease among individuals with BWSp.

Evolution over Time of Leg Length Discrepancy in Patients with Syndromic and Isolated Lateralized Overgrowth

Diana Carli et al.

Summary: The study demonstrated that leg length discrepancy associated with isolated or syndromic lateralized overgrowth tends to worsen with growth and correlates with the discrepancy at first observation. Isolated lateralized overgrowth tends to have a milder evolution, while Beckwith-Wiedemann spectrum predisposes to a more severe outcome.

Summary: The study investigated the relationship between clinical features and molecular diagnostic results in Beckwith-Wiedemann syndrome (BWS) and found a low molecular diagnostic rate correlated with a low risk of embryonal tumors in individuals with isolated lateralized overgrowth (ILO).

Summary: Beckwith-Wiedemann Spectrum (BWSp) is a common epigenetic childhood cancer predisposition disorder, with tumor risk and phenotypic differences compared to Classic BWS. Certain phenotypic features may be associated with specific epigenetic causes, indicating a need for further research.

Lateralized and Segmental Overgrowth in Children

Alessandro Mussa et al.

Summary: Congenital lateralized or segmental overgrowth (LO) disorders are heterogeneous conditions characterized by excessive tissue growth in a body region, caused by DNA anomalies. The clinical presentation of LO is diverse, posing challenges in diagnosis and management, requiring specific cancer screening and treatment.

Prenatal features in Beckwith-Wiedemann syndrome and indications for prenatal testing

Diana Carli et al.

Summary: This study investigates the prenatal phenotype of patients with Beckwith-Wiedemann spectrum (BWSp), revealing a higher rate of assisted reproductive techniques (ART) conception and twinning. Biochemical screening tests during pregnancy showed elevated levels of alpha-fetoprotein, uEstriol, and total human chorionic gonadotrophin. Morphology ultrasound scans indicated enlarged abdominal and head circumferences, along with various other abnormalities in the fetuses.
