New concepts in EBV-associated B, T, and NK cell lymphoproliferative disorders

Leticia Quintanilla-Martinez et al.

Summary: EBV-associated lymphoproliferative disorders (LPD) encompass a wide range of clinicopathological conditions derived from B, T, and NK cells. Progress has been made in the classification and understanding of the biology of these disorders since the 2016 WHO lymphoma classification. The diagnostic criteria have been refined and a new category of EBV-positive polymorphic B cell LPD has been introduced. The differential diagnosis of these conditions is challenging, and this report provides criteria to assist pathologists in making a diagnosis.

Classification and diagnostic evaluation of nodal T- and NK-cell lymphomas

Andrew L. Feldman et al.

Summary: This article discusses the classification and diagnostic evaluation of nodal T- and NK-cell lymphomas based on the 2022 International Consensus Classification of Mature Lymphoid Neoplasms (2022 ICC). It highlights the grouping of T-follicular helper cell lymphoma into a single entity with three subtypes, as well as the genetic subtype of ALK-negative anaplastic large cell lymphoma (ALCL) associated with DUSP22 rearrangement. The introduction of a new provisional entity, primary nodal EBV-positive T-/NK-cell lymphoma, is also mentioned, along with evolving molecular data indicating distinct subgroups within PTCL, NOS.

Update on Molecular Diagnosis in Extranodal NK/T-Cell Lymphoma and Its Role in the Era of Personalized Medicine

Ka-Hei (Murphy) Sun et al.

Summary: Natural killer (NK)/T-cell lymphoma (NKTCL) is an aggressive malignancy with unique characteristics. Research has found that its molecular mechanisms mainly involve chromosome deletion, JAK-STAT, PD-1/PD-L1 pathways, and mutations in tumor suppressor genes. These findings could influence traditional chemoradiotherapy and provide prospects for molecular diagnosis and treatment of NKTCL.

The Pathologic and Genetic Characteristics of Extranodal NK/T-Cell Lymphoma

Hyunsung Kim et al.

Summary: Extranodal NK/T-cell lymphoma is a tumor that occurs in extranodal sites, most commonly in the nasal cavity. It is associated with the presence of the EBV genome, specific immunophenotypes, and mutations in certain pathways and genes. Understanding the molecular pathogenesis of ENKTL and developing better treatment strategies are important for managing this disease.

Gehong Dong et al.

Summary: The study utilized two different CRISPR/Cas9 approaches to generate PRDM1-/- primary NK cells to investigate the role of PRDM1 in NK cell homeostasis. Results showed that PRDM1(-/-) NK cells exhibited higher proliferation rate and lower apoptosis compared to wild-type cells, with gene expression profiling revealing enrichment in pathways associated with proliferation and cell cycle.

Summary: The Epstein-Barr virus (EBV) is a common virus infecting the majority of individuals early in life, but only a minority develop EBV-related diseases under conditions of immunosuppression. EBV-driven lymphoid proliferations can originate from B-cells or T/NK-cells, with behaviors ranging from indolent to aggressive, posing challenges for diagnosis and treatment.

What Do We Have to Know about PD-L1 Expression in Prostate Cancer? A Systematic Literature Review. Part 1: Focus on Immunohistochemical Results with Discussion of Pre-Analytical and Interpretation Variables

Andrea Palicelli et al.

Summary: Immune checkpoint therapy targeting the PD-1-PD-L1 axis has shown promising results in treating immunologically ''hot'' tumors, with pembrolizumab demonstrating good therapeutic activity in selected prostate cancer patients. Analysis of PD-L1 expression in human prostate cancer samples revealed varying positivity rates across different histological subtypes, with different PD-L1 antibody clones showing differential expression patterns in tumor cells. Tumor heterogeneity, inter-institutional preanalytics, and variability in interpretation may lead to biases in results.

EBV-positive B-cell lymphomas and lymphoproliferative disorders: Review from the perspective of immune escape and immunodeficiency

Akira Satou et al.

Summary: The Epstein-Barr virus (EBV) is detected in a variety of B-cell lymphomas (BCLs) and B-cell lymphoproliferative disorders (B-LPDs), with immunodeficiency and immune escape playing key roles in their pathogenesis. The PD-1/PD-L1 pathway is crucial for immune escape, and PD-L1 immunohistochemistry (IHC) is a useful method for predicting immune escape mechanisms in lymphoid malignancies. Immune checkpoint inhibitors have shown promise in treating lymphomas characterized by immune escape, making them good candidates for PD1/PD-L1 blockade therapy.
