Small junction, big problems: Neuromuscular junction pathology in mouse models of amyotrophic lateral sclerosis (ALS)

Abrar Alhindi et al.

Summary: Amyotrophic lateral sclerosis (ALS) is a motor neuron disease with a heterogeneous clinical and genetic phenotype, and mouse models have shown variable disease progression at the neuromuscular junction (NMJ). Models with SOD1, TARDBP, and FUS mutations exhibit early NMJ dysfunction as a common factor, while C9ORF72 models do not follow the same pattern. Further study into the timing of NMJ pathology in newer ALS mouse models will confirm its pivotal role in ALS pathogenesis.

Melatonin promotes regeneration of injured motor axons via MT1 receptors

Marco Stazi et al.

Summary: Melatonin, a hormone produced by various tissues, has been found to accelerate nerve repair by activating the MT1 receptor and sustaining the ERK1/2 pathway. This receptor-mediated, pro-regenerative action of melatonin suggests its potential as a safe candidate for treating peripheral neurodegenerative conditions.

Summary: Successful regeneration of the neuromuscular junction relies on two fundamental steps: proper arrival of regenerating axons to muscle domains and resilience of postsynaptic domains in morphological and functional terms.

Expanding the Toolkit for In Vivo Imaging of Axonal Transport

Andrew P. Tosolini et al.

Summary: This paper details protocols for assessing in vivo axonal transport in mice, including distinguishing between motor and sensory neurons to analyze different neuronal subtypes, and evaluating the transport of signaling endosomes and mitochondria simultaneously. These complementary intravital approaches facilitate quantitative monitoring of axonal transport in health and disease by imaging different cargoes in distinct peripheral nerve axons.

Small Molecule Inhibitors of TEAD Auto-palmitoylation Selectively Inhibit Proliferation and Tumor Growth of NF2-deficient Mesothelioma

Tracy T. Tang et al.

Summary: Mutations in the NF2 gene leading to dysfunctional Merlin are common in malignant mesothelioma. Compounds selectively inhibiting YAP/TAZ-TEAD gene transcription have been discovered, providing a novel way to target the dysregulated Hippo-YAP pathway in mesothelioma and other YAP-driven cancers and diseases.
