Siglec receptors as new immune checkpoints in cancer

Michal A. Stanczak et al.

Summary: Cancer immunotherapy has shown promising results in improving the treatment and prognosis of patients, but resistance to current therapies remains a challenge. This review explores the potential of Siglec receptors as new immune checkpoints and targets for cancer immunotherapy. The authors summarize experimental evidence, discuss mechanisms of action, and highlight ongoing efforts to target Siglec receptors and their interactions with sialic acid Siglec-ligands.

Taxanes in cancer treatment: Activity, chemoresistance and its overcoming

Luciana Mosca et al.

Summary: Taxanes have been widely used as microtubule-targeting antitumor agents since 1984, showing impacts on key molecular mechanisms and cellular processes in cancer treatment. Research into the diverse effects and molecular mechanisms of taxanes may lead to more rational and efficacious applications, as well as novel strategies to overcome drug resistance in cancer therapy.

Single-cell analyses reveal key immune cell subsets associated with response to PD-L1 blockade in triple-negative breast cancer

Yuanyuan Zhang et al.

Summary: The study indicates that high levels of baseline CXCL13(+) T cells are associated with effective responses to combination therapy in triple-negative breast cancer patients. These cells increase following combination therapy but decrease after monotherapy, highlighting their importance in predicting treatment outcomes.

Dendritic Cell Regulation of T Helper Cells

Xiangyun Yin et al.

Summary: Dendritic cells play a crucial role in sensing the microenvironment and shaping the adaptive immune response. They consist of several subsets that regulate T lymphocyte differentiation and determine distinct CD4(+) T helper cell differentiation outcomes. The intrinsic properties of DC subsets, local tissue microenvironments, and interactions with other immune cells contribute to Th cell differentiation during homeostasis and inflammation.

Preoperative pembrolizumab combined with chemoradiotherapy for oesophageal squamous cell carcinoma (PALACE-1)

Chengqiang Li et al.

Summary: This study investigated the safety and efficacy of preoperative pembrolizumab combined with chemoradiotherapy in resectable oesophageal squamous cell carcinoma patients. The results showed that this treatment was safe and induced a pathological complete response in 55.6% of the tumors.

Chemotherapeutic and targeted agents can modulate the tumor microenvironment and increase the efficacy of immune checkpoint blockades

Jun-Yan Li et al.

Summary: The development of immune checkpoint blockade-based immunotherapy has changed cancer treatment methods significantly, although not all patients can benefit from it. Conventional chemotherapy and targeted therapy can enhance antitumor immune responses, providing a theoretical basis for combination therapy with ICBs, but the optimal treatment regimens still need further exploration.

Integrated single-cell transcriptome analysis reveals heterogeneity of esophageal squamous cell carcinoma microenvironment

Huy Q. Dinh et al.

Summary: This study utilized single-cell RNA sequencing to characterize the heterogeneous microenvironment of ESCC, identifying a tumor-specific subset of CST1(+) myofibroblasts with prognostic values and immunosuppressive features. The analysis of stromal cell types in the ESCC microenvironment provides a rich resource for further understanding of ESCC biology.
