Scanning structural mapping at the Life Science X-ray Scattering Beamline

Lin Yang et al.

Summary: This work introduces the instrumentation and software used for microbeam scattering and structural mapping at the Life Science X-ray Scattering beamline at NSLS-II. The adjustable beam size and simultaneous collection of scattering data at small and wide angles are achieved using a two-stage focusing scheme and multiple Pilatus detectors. Fly scans are implemented to minimize time interval between scattering patterns, and an in-vacuum sample environment is developed to reduce background scattering. The processing of data involves quantifying features from reciprocal coordinates and constructing structural maps. The paper presents examples of mapping onion epidermal cell walls and tomographic sectioning of an intact poplar stem.

Systems biology-guided understanding of white-rot fungi for biotechnological applications: A review

Teeratas Kijpornyongpan et al.

Summary: Plant-derived biomass is a rich carbon source on Earth, and white-rot fungi have the unique ability to efficiently break down plant cell walls. They play a crucial role in global carbon cycling and have potential applications in biotechnology. Current research on white-rot fungi has mainly focused on their extracellular enzymes, while their intracellular metabolism remains less explored. Systems biology is a powerful tool to study the biological processes of white-rot fungi.

Intracellular pathways for lignin catabolism in white-rot fungi

Carlos del Cerro et al.

Summary: Research shows that two white-rot fungi can utilize lignin as a carbon source and funnel carbon from lignin-derived aromatic compounds into central carbon metabolism via intracellular catabolic pathways. These insights into global carbon cycling and the foundation for a sustainable bioeconomy highlight the importance of simultaneous lignin depolymerization and bioconversion by white-rot fungi.

Towards an Understanding of Oxidative Damage in an α-L-Arabinofuranosidase of Trichoderma reesei: a Molecular Dynamics Approach

Jesus D. Castano et al.

Summary: Research on Trichoderma reesei revealed that the important enzyme alpha-L-arabinofuranosidase is susceptible to oxidative damage during industrial processing, leading to loss of function. Proteomics and molecular dynamics simulations showed significant changes in the conformational stability of the enzyme and its interactions with ligands when oxidized. The results provide insights into the molecular basis of enzyme inactivation in the commercially relevant fungus T. reesei.

Summary: Among 40 fungal strains tested, Byssochlamys nivea MUT 6321 exhibited the best growth performance with different concentrations of inhibitors, being the only fungus capable of growing when all four molecules were present simultaneously. It was able to completely degrade furfural within 24 hours and phenolic aldehydes in less than 11 days, showing potential for removing toxic compounds from pretreated lignocellulosic biomass.
