Multimodal regulatory elements within a hormone-specific super enhancer control a heterogeneous transcriptional response

Summary: The hormone-stimulated glucocorticoid receptor (GR) interacts with enhancers and GR binding sites (GBSs) to modulate transcription. This study found that hormone treatment affects enhancer dynamics in breast cancer cells. Additionally, it was discovered that different GBSs have unique roles in the hormone response, influencing transcription in different ways.

Canonical WNT signaling-dependent gating of MYC requires a noncanonical CTCF function at a distal binding site

Summary: The gene gating of MYC oncogenic super-enhancer plays an important role in colon cancer cells, but its mechanism is poorly understood. This study reveals that WNT signaling increases MYC expression in colon cancer cells by facilitating nuclear export of MYC mRNA through the regulation of CTCF binding site within the oncogenic super-enhancer. Furthermore, it is found that the trafficking of the oncogenic super-enhancer to the nuclear pore is essential for the growth advantage of cancer cells.

Systematic comparison of CRISPR-based transcriptional activators uncovers gene-regulatory features of enhancer-promoter interactions

Summary: This study systematically compared different dCas9-based transcriptional activators and found that their expression levels and transactivation efficacies vary in different human cell types. The study also showed that these activators can induce the production of enhancer RNAs, which are positively correlated with downstream mRNA expression. Additionally, the study demonstrated the existence of intrinsic transcriptional and epigenetic reciprocity between human enhancers and promoters.

Comprehensive characterization of enhancer RNA in hepatocellular carcinoma reveals three immune subtypes with implications for immunotherapy

Summary: In this study, the genome-wide eRNA landscape in HCC was revealed, and individuals with HCC were divided into three immune-related clusters based on eRNA expression profiles. The results showed that C1 cluster had enriched immune infiltration and may be sensitive to immune checkpoint inhibitors. C2 cluster exhibited immune depletion characteristics, malignant clinical features, and the worst prognosis, suggesting potential benefits from targeted therapy. C3 cluster presented moderate immune infiltration, metabolism-related signatures, and the best prognosis. A 51-eRNA classifier was constructed for subtype prediction and validated in two independent cohorts.
