Comparison and evaluation of statistical error models for scRNA-seq

Saket Choudhary et al.

Summary: This study analyzes 59 scRNA-seq datasets to evaluate the performance of different error models in modeling variation in scRNA-seq data. The results show clear evidence of overdispersion for genes with sufficient sequencing depth, necessitating the use of a negative binomial model. The degree of overdispersion varies widely across datasets, systems, and gene abundances.

Peculiar protrusions along tanycyte processes face diverse neural and nonneural cell types in the hypothalamic parenchyma

Roxane Pasquettaz et al.

Summary: The study investigates the neuroanatomical associations between tanycytes and neural cells, particularly in the arcuate nucleus. Tanycytes were found to have subcellular protrusions contacting different neural cells, including NPY and POMC neurons in the arcuate nucleus. The protrusions were observed to contain ribosomes, mitochondria, vesicles, and transporters, indicating dense communications between tanycytes, neurons, and blood vessels.

What is the physiological role of hypothalamic tanycytes in metabolism?

Matei Bolborea et al.

Summary: Tanycytes, peculiar cells in the metabolic hypothalamus, play a multifunctional role in controlling energy balance. Their physiological function in maintaining metabolic homeostasis is still unclear, leading to ambiguity in the field and providing new research directions. Furthermore, the potential involvement of tanycytes in metabolic disorders and age-related neurodegenerative diseases will be discussed.

Ablation of glucokinase-expressing tanycytes impacts energy balance and increases adiposity in mice

Antoine Rohrbach et al.

Summary: This study investigated the role of GCK-expressing tanycytes in the regulation of energy balance in mice. The results showed that Gck deletion induced the death of tanycytes and affected adiposity in mice, as well as NPY neuron function.

Control of neurogenic competence in mammalian hypothalamic tanycytes

Summary: Disruption of the NFI family of transcription factors stimulates tanycyte proliferation and neurogenesis in the postnatal hypothalamus. NFI factors repress Shh and Wnt signaling in tanycytes, controlling proliferation and neurogenesis. Tanycytes deficient in Nfia/b/x give rise to various hypothalamic neuronal subtypes, suggesting potential for remodeling hypothalamic neural circuitry.
