Hypoxia-Preconditioned Extracellular Vesicles from Mesenchymal Stem Cells Improve Cartilage Repair in Osteoarthritis

Summary: This study aimed to compare the effects of hypoxia-preconditioned and normoxia-preconditioned MSC-EVs on OA cartilage repair, as well as to investigate the impact of hypoxia on microRNA expression in MSC-EVs. The results demonstrated that hypoxia-preconditioned MSC-EVs exhibited superior effects in enhancing chondrocyte proliferation and migration and suppressing chondrocyte apoptosis compared to normoxia-preconditioned MSC-EVs. Additionally, bioinformatics analysis revealed that hypoxia may promote cartilage repair through the miRNA-18-3P/JAK/STAT or miRNA-181c-5p/MAPK signaling pathway.

Nrf2-mediated anti-inflammatory polarization of macrophages as therapeutic targets for osteoarthritis

Summary: Macrophages in synovial joints play an important role in the pathological process of osteoarthritis (OA). Their transition between pro-inflammatory and anti-inflammatory phenotypes can impact the intra-articular microenvironment. The inhibition of NF-kappa B signaling pathway is crucial in OA treatment, while the activation of Nrf2 signaling pathway, which has antioxidant and anti-inflammatory effects, and its crosstalk with NF-kappa B pathway have received less attention. Activating Nrf2 can inhibit M1 polarization and promote M2 polarization through multiple signaling pathways. Various potential candidates for activating Nrf2 to promote M2 phenotype macrophages in OA treatment are discussed.
