Development of Organoids from Mouse Pituitary as In Vitro Model to Explore Pituitary Stem Cell Biology

Emma Laporte et al.

Summary: Researchers have developed a new and innovative in vitro organoid model to study the biology of pituitary stem cells. This model allows for the development of 3D cell structures that closely resemble the features of stem cells and their tissues, providing a powerful tool for studying key conditions of pituitary remodeling.

Pituitary stem cells produce paracrine WNT signals to control the expansion o their descendant progenitor cells

John P. Russell et al.

Summary: Research has shown that pituitary stem cells play a significant role in pituitary growth and renewal, with SOX2(+) stem cells identified as a key source of WNT ligands. By blocking the secretion of WNTs from SOX2(+) stem cells, the proliferation of nearby committed progenitor cells decreases, demonstrating the dependence of progenitor cell multiplication on paracrine WNT signaling from stem cells.

Pituitary Remodeling Throughout Life: Are Resident Stem Cells Involved?

Emma Laporte et al.

Summary: The pituitary gland has the ability to dynamically adapt its cell composition to changing hormonal needs throughout life. Different hormonal cell populations expand during growth and development, and decline in function is associated with aging. The involvement and phenotype of pituitary stem cells in pituitary remodeling events remain largely unknown, presenting unanswered questions in pituitary research.

Interleukin-6 is an activator of pituitary stem cells upon local damage, a competence quenched in the aging gland

Annelies Vennekens et al.

Summary: The study identifies IL-6 as a pituitary stem cell activator upon local damage, with the competence quenched at aging. In aging pituitary, cells exhibit elevated basal IL-6 levels and an inflammatory state, inhibiting the activation response to IL-6. Interestingly, in vitro stem cell activation by IL-6 was observed in organoid culture not only from young but also from aging pituitary.
