Surgical strategy of locally advanced differentiated thyroid cancer

Keisuke Enomoto et al.

Summary: Approximately 20% of patients with differentiated thyroid cancer (DTC) have direct tumor extension with invasion of surrounding tissues. Molecular-targeted therapy has shown improvements in survival outcome, but has also presented fatal adverse events in locally-advanced DTC. Surgery still plays an important role in DTC management, especially in local control. This review article examines the surgical strategy for advanced DTC.

Lipocalin-2 silencing suppresses inflammation and oxidative stress of acute respiratory distress syndrome by ferroptosis via inhibition of MAPK/ERK pathway in neonatal mice

Xiaodong Wang et al.

Summary: This study suggests that LCN2 plays an important regulatory role in neonatal ARDS by inhibiting ferroptosis-related inflammation and oxidative stress through the inhibition of the MAPK/ERK pathway, thus exerting a protective effect.

Advanced RAI-refractory thyroid cancer: an update on treatment perspectives

Olga Karapanou et al.

Summary: The knowledge on molecular biology of follicular cell-derived thyroid cancer has led to the development of novel therapies, particularly tyrosine kinase inhibitors. These targeted therapies have changed treatment strategies and disease prognosis, but drug resistance remains a major challenge. Understanding the molecular pathways and escape mechanisms of the tumor is crucial for the development of more effective drugs for thyroid cancer.

Association between Gene Promoter Methylation and Cervical Cancer Development: Global Distribution and A Meta-analysis

Aissam El Aliani et al.

Summary: DNA methylation is a key epigenetic event related to gene silencing and carcinogenesis. The meta-analysis evaluated the association between promoter methylation of specific genes and the risk of cervical cancer development, finding higher methylation frequencies in certain genes in cervical cancer cases compared to controls. Promoter methylation could potentially serve as a promising noninvasive biomarker for early cervical lesions.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Delineation of colorectal cancer ligand-receptor interactions and their roles in the tumor microenvironment and prognosis

Hexin Lin et al.

Summary: This study investigated the patterns of ligand-receptor interactions in colorectal cancer (CRC) and their effects on the tumor microenvironment, immunotherapy, and prognosis of patients. By analyzing over 55,000 single-cell RNA sequencing samples, the study identified tumor subtypes based on LRIs, explored the relationship between subtypes and immunotherapy efficacy, and discovered specific ligand-receptor pairs affecting tumor-infiltrating lymphocytes. Finally, a prognostic model based on ligand-receptor pairs was developed and validated, providing new insights for CRC subtype classification, risk screening, and potential therapeutic targets.

Blockade of mutant RAS oncogenic signaling with a special emphasis on KRAS

Robert Roskoski

Summary: RAS proteins play crucial roles in physiological signal transduction processes related to cell growth, division, and survival. RAS mutations, especially KRAS mutations, are common in various cancers, with potential implications for targeted therapy. Covalent modification of the KRAS C12 led to the discovery of a new pocket, facilitating the development of second-line treatment for KRAS(G12C)-mutant non-small cell lung cancer. Efforts are also being made to develop inhibitors targeting MAP kinase and PI3-kinase pathways as indirect RAS antagonists.

miR-486 Promotes the Invasion and Cell Cycle Progression of Ovarian Cancer Cells by Targeting CADM1

Chenyang Li et al.

Summary: Our study demonstrated that miR-486 is significantly upregulated in ovarian cancer tissues and cells, while CADM1 expression is downregulated. miR-486 targets CADM1, and interference with miR-486 can inhibit cell proliferation and invasion while promoting apoptosis. Knocking down both miR-486 and CADM1 enhances cell proliferation and invasion, shifts cell cycle progression, and reduces apoptosis in SKOV3 cells.

Focal adhesion kinase inhibitors, a heavy punch to cancer

Yueling Wu et al.

Summary: Kinases are ideal druggable targets for diseases, especially in cancer therapy. Focal adhesion kinase (FAK), a non-receptor tyrosine kinase, plays a significant role in cancer progression, with FAK inhibitors showing promising results in preclinical and early-stage clinical trials. Targeting FAK in combination strategies may be more effective in reversing the failure of chemotherapies or targeted therapies in solid tumors.

Endoplasmic reticulum stress signals in the tumour and its microenvironment

Xi Chen et al.

Summary: This review explores how ER stress in the tumor microenvironment can influence the functions of cancer cells and immune cells, leading to vulnerabilities that could be targeted by emerging therapeutic strategies. The tightly regulated processes of protein handling, modification, and folding in the ER determine cell function and survival, and disruptions in ER homeostasis can result in a state of persistent ER stress with pro-tumoral effects in cancer cells and immunomodulatory effects in immune cells.
