A global view of the interplay between non-alcoholic fatty liver disease and diabetes

Norbert Stefan et al.

Summary: Non-alcoholic fatty liver disease (NAFLD) has become an epidemic, associated with the increasing prevalence of obesity and type 2 diabetes, and its pathophysiology is closely related to the development of communicable diseases such as COVID-19. Understanding the relationship between diabetes and NAFLD is important for disease treatment. Identifying different pathological mechanisms of NAFLD among different groups of diabetes patients may improve diagnosis and prediction.

Glutaminolysis-ammonia-urea Cycle Axis, Non-alcoholic Fatty Liver Disease Progression and Development of Novel Therapies

Angela Rojas et al.

Summary: The prevalence of non-alcoholic fatty liver disease (NAFLD) is increasing worldwide, and its development is closely linked to obesity, insulin resistance, type 2 diabetes mellitus, and metabolic syndrome. The cause of NAFLD-linked fibrosis progression is still unclear, posing challenges for the development of new therapies. Cirrhosis is associated with the activation of hepatic stellate cells (HSC) and excess accumulation of extracellular matrix proteins. Oxidative stress and glutaminolysis are potential antifibrotic targets. This review focuses on the role of the glutaminolysis-ammonia-urea cycle axis in NAFLD progression and suggests it as a novel therapeutic target.

Summary: This study investigated alcohol consumption in patients with presumed NAFLD and ALD, using novel objective alcohol markers. The results showed that approximately 28.6% of presumed NAFLD patients and 25% of MAFLD patients are at risk of alcohol-related liver damage.

Liver injury in non-alcoholic fatty liver disease is associated with urea cycle enzyme dysregulation

Rocio Gallego-Duran et al.

Summary: This study aimed to evaluate changes in urea cycle enzymes in NAFLD patients and animal models, and found that liver fibrosis and steatosis were associated with a reduction in both gene and protein expression patterns of mitochondrial urea cycle enzymes.

Pinar Gokcen et al.

Summary: Research showed that lactulose and polyethylene glycol 3350, at non-diarrheal doses, reduced hepatic inflammation in mice with induced NAFLD. Polyethylene glycol 3350 also decreased hepatic steatosis and body weight.

Spatio-Temporal Multiscale Analysis of Western Diet-Fed Mice Reveals a Translationally Relevant Sequence of Events during NAFLD Progression

Summary: In this study, time-resolved characterization of a mouse model of NAFLD identified a sequence of eight key events in the development of the disease, along with functional changes and transcriptomic landscape similarities to human NAFLD and HCC. The findings provide a basis for identifying therapeutic targets in NAFLD.
