Metal-free synthesis of gem-difluorinated heterocycles from enaminones and difluorocarbene precursors

Summary: A cascade strategy for synthesizing gem-difluorinated 2H-furans from the reactions of BrCF2CO2Et with enaminones has been described. The reactions can tolerate a wide range of functional groups under metal-free conditions, and an active aminocyclopropane is proposed as a key intermediate for the synthesis of gem-difluorinated 2H-furans.

Iron-catalyzed hydroaminocarbonylation of alkynes: Selective and efficient synthesis of primary α,β-unsaturated amides

Summary: A Rh(III)-catalyzed dehydrogenative annulation and spirocyclization of 2-arylindoles and 2-(1H-pyrazol-1-yl)-1H-indole with maleimides has been developed, providing highly functionalized compounds. The reaction methodology has broad substrate scope, good functional group tolerance, and is simple and scalable. The annulated product of 2-(1H-pyrazol-1-yl)-1H-indole showed significant red-shift and high absorption and emission values compared to 2-phenylindole.

A Strategy for Amide C-N Bond Activation with Ruthenium Catalyst: Selective Aromatic Acylation

Summary: This study introduces a strategy for amide C-N bond activation using a ruthenium catalyst, where in situ formed bis-cycloruthenated complexes are demonstrated as key active species with superior oxidative addition ability. The direct C-H bond activation of 2-arylpyridines followed by amide C-N bond activation in the presence of a ruthenium precatalyst resulted in moderate to good yields of monoacylation products, with synthetically useful functional groups remaining intact during the tandem C-H/C-N bond activation reactions.

The developed protocol demonstrated excellent yields and versatility, and was further extended to synthesize Trypanosome brucie and luciferase inhibitors.

UV-Light-Induced N-Acylation of Amines with α-Diketones
