The ALDH Family Contributes to Immunocyte Infiltration, Proliferation and Epithelial-Mesenchymal Transformation in Glioma

Zeyu Wang et al.

Summary: In this study, the role of the aldehyde dehydrogenase family in gliomas was explored through bulk RNA-seq analysis and single-cell RNA-Seq analysis. The results showed that high-scoring samples based on the aldehyde dehydrogenases family were associated with worse survival outcomes. Pathway enrichment scores were calculated to validate the biofunction prediction results, indicating that the aldehyde dehydrogenases family influenced glioma progression by modulating tumor cell proliferation, migration, and immune landscape. ALDH3B1 and ALDH16A1, two main contributors to the scoring system, were found to affect glioblastoma cell proliferation and migration through cell-cycle arrest and the epithelial-mesenchymal transition.

Vidhya M. Ravi et al.

Summary: This study utilizes spatially resolved transcriptomics, metabolomics, and proteomics to characterize glioblastomas and uncover their spatial organization and adaptive mechanisms to inflammatory and metabolic stimuli. The integration of metabolic imaging and imaging mass cytometry reveals the interdependence between tumor and host in a locoregional manner, leading to spatially exclusive adaptive transcriptional programs. The findings confirm the importance of environmental stress in shaping glioblastoma subclones and their transcriptional programs.

Frederick S. Varn et al.

Summary: This study analyzed RNA and DNA sequencing data from tumor pairs of patients with IDH-wild-type and IDH-mutant glioma, and identified the factors driving therapy resistance and recurrence. The results showed that IDH status, histological features, somatic alterations, and microenvironment interactions play important roles in glioma recurrence. Furthermore, specific cellular and genetic changes were associated with tumor growth, invasion, and neuronal interactions, providing potential targets for altering disease progression.

Characterization of Different Subtypes of Immune Cell Infiltration in Glioblastoma to Aid Immunotherapy

Peng Feng et al.

Summary: Glioblastoma multiforme (GBM) is a frequently occurring adult primary brain cancer with high aggressiveness. Immunotherapy has potential in treating GBM, but the complex tumor microenvironment poses challenges. In this study, different subtypes of immune cell infiltration were identified through clustering analysis, and gene cluster A with a hot immune cell infiltration phenotype was found to have a better prognosis and improved immunotherapy response.

Glioblastomas acquire myeloid-affiliated transcriptional programs via epigenetic immunoediting to elicit immune evasion

Ester Gangoso et al.

Summary: In this study, it was found that GBM stem cells acquire the ability to evade immune clearance through epigenetic immunoediting, leading to the formation of an immunosuppressive tumor microenvironment. This process is not driven by genetic selection but by altering the transcriptional and epigenetic profile of GSCs after immune attack.

Development of a T-cell activation-related module with predictive value for the prognosis and immune checkpoint blockade therapy response in glioblastoma

Zihao Yan et al.

Summary: This study established a T-cell activation-related risk score based on mRNA expression profiles to evaluate the prognosis and immunotherapy response of GBM patients. The high-risk group had poorer prognosis, stronger immune response, and a more complex tumor immune microenvironment. The immunotherapy score obtained by combining the risk and priming scores was found to be a good evaluation index for ICB response in GBM patients, providing potential treatment opportunities.

Exploring the longitudinal glioma microenvironment landscape uncovers reprogrammed pro-tumorigenic neutrophils in the bone marrow

Prerna Magod et al.

Summary: Recent multi-omics studies have shown different compositions of the immune tumor microenvironment (TME) in glioblastoma (GBM). This study focused on the cellular evolution of the TME in murine GBM models initiated from different oncogenic drivers. It revealed that neutrophils infiltrate early during tumor progression, primarily in the mesenchymal GBM model. Depletion of neutrophils at the onset of disease was found to accelerate tumor growth and decrease mouse survival time, indicating a potential point of intervention in GBM treatment.
