Optimization of topical formulations using a combination of in vitro methods to quantify the transdermal passive diffusion of drugs

Joanna Hummer et al.

Summary: This paper presents a new approach to optimize topical products and select lead formulation candidates. The study compares the diffusion of diclofenac through all skin layers using open flow microperfusion and the Franz diffusion cell. The results demonstrate that the diffusion of diclofenac through the dermis and subcutis does not correlate with its diffusion through the skin barrier, and the Franz diffusion cell data alone cannot predict it. This approach provides a convenient tool for product development and selection.

Atopic dermatitis: an expanding therapeutic pipeline for a complex disease

Thomas Bieber

Summary: Recent advances in understanding of the complex phenotype and mechanisms underlying atopic dermatitis (AD) have revealed multiple new potential targets for pharmacological intervention. There are now more than 70 new compounds in development, in addition to regulatory approval for several inhibitors in Europe. The potential for a precision medicine approach to enable prevention and more effective long-term control of this complex disease is highlighted.

Summary: This global survey in 18 countries revealed that atopic dermatitis affects a substantial proportion of the pediatric population, with less than 15% having severe cases. Prevalence and severity varied across countries and age groups, with rural areas generally having lower prevalence than urban or suburban areas.

Cost-of-illness of atopic dermatitis in Germany: data from dermatology routine care

N. Mohr et al.

Summary: The study aimed to determine the annual costs of treating adult patients with AD in Germany, finding the total economic burden for treating adult patients with AD in Germany estimated at more than 2.2 billion euros, and indicating that AD has a moderate level of costs-of-illness in Germany compared to conditions like psoriasis. The approval of new therapies is expected to lead to markedly more resource utilization in the next years.

A Proinflammatory Cytokine Network Profile in Th1/Type 1 Effector B Cells Delineates a Common Group of Patients in Four Systemic Autoimmune Diseases

Quentin Simon et al.

Summary: This study found that a proinflammatory cytokine profile characterized by high levels of CXCL10, interleukin-6 (IL-6), IL-2, and tumor necrosis factor defined a distinct group of patients in four systemic autoimmune diseases. This proinflammatory cluster was associated with specific circulating immune cell signature in each disease, more severe disease, and higher levels of autoantibodies, suggesting an uncontrolled proinflammatory Th1 immune response.

Gut-Skin Axis: Current Knowledge of the Interrelationship between Microbial Dysbiosis and Skin Conditions

Britta De Pessemier et al.

Summary: The microbiome plays a crucial role in various skin disorders, with dysbiosis in the skin and gut microbiome associated with altered immune responses that contribute to the development of skin diseases. Enhanced understanding of the microbiome's involvement in human skin diseases and its interaction with diet and immune responses is essential for developing new therapeutic approaches.

The power and potential of BIOMAP to elucidate host-microbiome interplay in skin inflammatory diseases

Harri Alenius et al.

Summary: Atopic dermatitis and psoriasis are two common chronic inflammatory skin diseases, and the underlying causes and mechanisms are still poorly understood. The BIOMAP project aims to investigate these diseases and identify potential biomarkers responsible for variations in disease outcomes. By integrating patient cohorts, data, and knowledge, the project has the potential to bridge the gap between current problems and solutions.
