Exosomes Recovered From the Plasma of COVID-19 Patients Expose SARS-CoV-2 Spike-Derived Fragments and Contribute to the Adaptive Immune Response

Elisa Pesce et al.

Summary: This study compared exosomes recovered from the plasma of MILD or SEVERE COVID-19 patients and found that exosomes from MILD patients efficiently regulated immune responses, while those from SEVERE patients were associated with increased and chronic inflammation.

Human skin-resident host T cells can persist long term after allogeneic stem cell transplantation and maintain recirculation potential

Gustavo P. de Almeida et al.

Summary: Tissue-resident memory T cells (T-RM) play crucial roles in host defense in various tissues and barrier sites. However, the maintenance and regulatory checkpoints of human T-RM cells are still poorly understood. A study on patients receiving allogeneic hematopoietic stem cell transplantation (allo-HSCT) revealed long-term sequestration of host T cells in human skin. This finding sheds light on the characteristics and functions of skin-resident T-RM cells in humans.

Human resident memory T cells exit the skin and mediate systemic Th2-driven inflammation

Johanna Strobl et al.

Summary: Patients with active GVHD have circulating cTRMs originating from the skin, which can produce pro-inflammatory Th2/Th17 cytokines and mediate keratinocyte damage, indicating their potential to propagate inflammation.

GVHD Prophylaxis 2020

Mahasweta Gooptu et al.

Summary: GVHD, both acute and chronic, is a major complication of allogeneic transplantation, and its prevention is crucial for the success of transplants. Current standards revolve around calcineurin inhibitors, but promising translational advances targeting T-cell regulation and co-stimulatory pathways are emerging. These approaches are based on a pathophysiology perspective and show potential for transforming GVHD prophylaxis in the near future.

Graft-versus-host disease: a disorder of tissue regeneration and repair

Ronjon Chakraverty et al.

Summary: In summary, regenerative failure at barrier surfaces and maladaptive repair leading to fibrosis are hallmarks of graft-versus-host disease (GVHD). While immunosuppressive treatment can control inflammation, impaired tissue homeostasis results in prolonged organ damage and impaired quality of life. Current interventions aim to address these critical failures in tissue regeneration and repair to improve therapeutic outcomes in GVHD.
