Activation of UQCRC2-dependent mitophagy by tetramethylpyrazine inhibits MLKL-mediated hepatocyte necroptosis in alcoholic liver disease

Summary: This study demonstrates that tetramethylpyrazine (TMP) extracted from Ligusticum Wallichii Franch has the potential to limit alcoholic liver disease-induced hepatocyte necroptosis. The mechanism involves the reduction of receptor-interacting protein kinase 1 (RIPK1)/RIPK3 necrosome activation and phosphorylation of mixed-lineage kinase domain-like protein (MLKL), inhibition of mitochondrial translocation of p-MLKL, disruption of the necroptotic signal loop, and promotion of mitophagy. Restoration of ubiquinol-cytochrome c reductase core protein 2 (UQCRC2) expression by TMP contributes to the inhibition of necroptosis. These findings suggest that TMP could be a promising therapeutic candidate for alcoholic liver disease.
