Overexpression of HIF-1 alpha is strongly associated with poor prognosis in several solid cancers. Pharmacological targeting of the HIF-1 signaling pathway has been considered a new strategy for cancer therapy, but translation into clinically available treatments remains challenging.

CircWHSC1 Promotes Breast Cancer Progression by Regulating the FASN/AMPK/mTOR Axis Through Sponging miR-195-5p

Summary: Studies have shown that circRNAs play a role in cancer progression. In this study, circWHSC1 was identified as a novel circRNA that accelerates ovarian cancer progression. The function of circWHSC1 in breast cancer was investigated, and it was found that circWHSC1 is up-regulated in breast cancer tissues and is associated with higher tumor stages, lymphatic metastasis, and worse survival of breast cancer patients. Functional experiments showed that circWHSC1 promotes breast cancer cell proliferation, migration, invasion, and tumor growth. Mechanistically, circWHSC1 functions as a competitive endogenous RNA by sponging miR-195-5p, which delays breast cancer progression. In addition, miR-195-5p regulates the expression of fatty acid synthase (FASN) and activates the AMPK/mTOR pathway. Inhibition of FASN or activation of the AMPK pathway reverses the oncogenic effects mediated by circWHSC1. Overall, circWHSC1 acts as an oncogene in breast cancer by modulating the miR-195-5p/FASN/AMPK/mTOR pathway.

This study used VEGFB gene-knockout mice to investigate the effects of downregulation of VEGFB on lipid metabolism and insulin secretion, and to explore the mechanism of VEGFB in the regulation of glucose and lipid metabolism. The findings suggest that downregulating VEGFB improves lipid metabolism and insulin resistance.
