Epidemiological, clinical and virological characteristics of four cases of monkeypox support transmission through sexual contact, Italy, May 2022

Summary: Since May 2022, there has been an ongoing monkeypox outbreak in non-endemic countries. Four cases in young adult men in Italy, who engaged in condomless sexual intercourse, were reported. The patients are in good clinical condition and do not require specific antiviral drugs. Monkeypox viral DNA was detected in seminal fluid samples. The possibility of sexual transmission of monkeypox virus needs to be investigated.

Emergence of multi-drug-resistant Mycobacterium tuberculosis in Niger: A snapshot based on whole-genome sequencing

Bioinformatic Mining and Structure-Activity Profiling of Baeyer-Villiger Monooxygenases from Mycobacterium tuberculosis

Summary: Tuberculosis (TB), caused by Mycobacterium tuberculosis, is a leading cause of global mortality and drug resistance is a rising problem in TB treatment, necessitating the development of innovative anti-TB drugs. Baeyer-Villiger monooxygenases (BVMOs) play a crucial role in the bioactivation of many anti-TB drugs, but their structural and functional features remain largely unknown. In this study, a comprehensive bioinformatic analysis confirmed the presence of six BVMOs in M. tuberculosis, including MymA, EthA, and Rv0565c, which are involved in the activation of the second-line prodrug ethionamide, as well as the novel BVMO Rv0892. Through a combination of in silico characterization and in vitro validation, we elucidated the structural framework and substrate preferences of these BVMOs. Particularly, MymA exhibited enhanced capacity and a distinct selectivity profile towards ligands, supporting the understanding of its specific catalytic site topology. These findings contribute to a better comprehension of the structure-function relationship in BVMOs and expand the repertoire of selective and/or overlapping BVMOs for improving anti-TB therapy.
