Development of a Freeze-Dried CRISPR-Cas12 Sensor for Detecting Wolbachia in the Secondary Science Classroom

Summary: Training the future synthetic biology workforce requires exposing students to biotechnology concepts and activities in secondary education. While polymerase chain reaction (PCR) is commonly used in classrooms for investigating and applying recombinant DNA technology, cutting-edge biotechnologies like CRISPR-based diagnostics have not been widely implemented. To address this gap, the authors present a freeze-dried CRISPR-Cas12 sensing reaction that complements traditional DNA technology education and facilitates the teaching of synthetic biology concepts. These reactions accurately detect Wolbachia bacteria in arthropod-derived PCR samples within two hours, can be stored for over a month without degradation, are easy to use, and cost less than $40 for a classroom of 22 students. The authors propose that incorporating these freeze-dried CRISPR-Cas12 reactions into the existing biology curriculum will expand educational opportunities in STEM fields.
