The pro-inflammatory response of macrophages regulated by acid degradation products of poly(lactide-co-glycolide) nanoparticles

Shufang Ma et al.

Summary: This study investigates the immunological effects of PLGA degradation products on cell viability, apoptosis, polarization, and inflammatory reaction. The results show that PLGA degradation induces an acid environment that decreases cell viability and increases cell apoptosis, impacting cell function. The degradation products of PLGA can also convert macrophages to a pro-inflammatory state, providing insight into PLGA-induced inflammation for the design of future biomaterials.

Summary: In this study, a PLGA scaffold combined with magnesium hydroxide and bone extracellular matrix was utilized to enhance anti-inflammation, mechanical properties, and osteoconductivity. Additionally, the development of a bioactive nanocomplex containing polydeoxyribonucleotide and bone morphogenetic protein 2 showed synergistic abilities in angiogenesis and bone regeneration, leading to outstanding performance in anti-inflammation, angiogenesis, and osteogenesis for the PME/NC scaffold. Such an advanced PME/NC scaffold suggests an integrated bone graft substitute for bone regeneration.

Amelioration of post-traumatic osteoarthritis via nanoparticle depots delivering small interfering RNA to damaged cartilage

Sean K. Bedingfield et al.

Summary: In this study, intra-articular injection of nanoparticles targeting collagen and delivering small interfering RNA for matrix metalloproteinase 13 effectively protected cartilage integrity and reduced disease progression in mice with osteoarthritis. The inhibition of MMP13 suppressed genes associated with tissue restructuring, angiogenesis, innate immune responses, and proteolysis. Targeting collagen in damaged extracellular matrix of arthritic cartilage may be an effective strategy for treating osteoarthritis.

The antagonistic effect of magnesium hydroxide particles on vascular endothelial activation induced by acidic PLGA degradation products

Kyoung-Won Ko et al.

Summary: This study demonstrated that PLGA degradation products accumulated in HCAECs through MCT1, leading to oxidative stress and the activation of the MAPK/NF-kappa B signaling pathway, resulting in vascular endothelial activation. However, the addition of MH particles significantly reduced this activation and inflammation-related factors expression, while Mg2+ released from MH particles restored endothelial function. These findings suggest that MH particles could prevent the accumulation of PLGA degradation products in HCAECs and suppress vascular endothelial activation.
