Targeted multicolor in vivo imaging over 1,000 nm enabled by nonamethine cyanines

Venu G. Bandi et al.

Summary: Through rational design, red-shifted, bioconjugatable heptamethine cyanine dyes were developed for multiplexed in vivo imaging in the shortwave-infrared/near-infrared-II region. The use of wavelengths between 1,000 and 2,000 nm enables high-resolution in vivo imaging at depths not possible with conventional optical wavelengths. Recent progress has shown a lack of bioconjugatable probes for imaging in the shortwave-infrared or near-infrared (NIR)-II range.

In vivo non-invasive confocal fluorescence imaging beyond 1,700 nm using superconducting nanowire single-photon detectors

Feifei Wang et al.

Summary: By studying light scattering and utilizing new materials technology, the imaging depth of high-resolution optical microscopy in live mammals can be improved. This study demonstrates single-photon excitation fluorescence imaging in the NIR-IIc range, achieving the longest single-photon excitation and emission wavelengths to date for in vivo imaging of mice.

Ultrafast two-photon fluorescence imaging of cerebral blood circulation in the mouse brain in vivo

Guanghan Meng et al.

Summary: Understanding blood flow dynamics is crucial for studying the function of blood vessels. Current methods lack the resolution to monitor blood flow at a cellular level in large vessels. However, using a fast line-scanning module, researchers were able to achieve high-resolution imaging of blood flow in the cortex of awake mice. This breakthrough allowed them to measure cerebral blood flow and observe pulsatile blood flow patterns.

In vivo NIR-II structured-illumination light-sheet microscopy

Feifei Wang et al.

Summary: The study developed a novel near-infrared II structured-illumination light-sheet microscopy for large volumetric three-dimensional imaging of deep tissues in live mammals, with improved spatial resolution and background suppression. This technology provides an additional tool for noninvasive volumetric molecular imaging of immune cells in live mammals.
