Placental diabesity: placental VEGF and CD31 expression according to pregestational BMI and gestational weight gain in women with gestational diabetes

Summary: The study investigated the placental expression of VEGF and CD31 in pregnancies complicated by gestational diabetes (GDM) and the influence of pregestational BMI and gestational weight gain (GWG) on this expression. The results showed that GDM pregnancies are characterized by increased placental expression of VEGF and CD31, and the expression of these markers is associated with maternal increased pregestational BMI and GWG.

Hepatic Suppression of Mitochondrial Complex II Assembly Drives Systemic Metabolic Benefits

Summary: Alternate day fasting (ADF) leads to metabolic reprogramming in the liver and dysfunction of complex II. Knockout of the Sdhaf4 gene in the liver improves glucose tolerance and systemic insulin sensitivity, similar to the effects of ADF intervention. Hepatocytes activate the arginine-nitric oxide (NO) biosynthesis axis in response to complex II and citric acid cycle dysfunction, and the released NO enhances insulin sensitivity in muscle and adipocytes.

β-Cell Succinate Dehydrogenase Deficiency Triggers Metabolic Dysfunction and Insulinopenic Diabetes

Summary: Mitochondrial dysfunction plays a central role in type 2 diabetes, and the pathogenic mechanisms in pancreatic beta-cells are not fully understood. In this study, using samples from diabetic individuals and a mouse model, the authors demonstrate that succinate dehydrogenase (SDH) deficiency drives mitochondrial dysfunction in beta-cell failure and insulinopenic diabetes. SDH deficiency impairs glucose-induced respiratory oxidative phosphorylation and compromises glucose-stimulated ATP production, insulin secretion, and beta-cell growth. The loss of SDH causes excess succinate accumulation, which activates metabolic anabolism and lipid synthesis. This study suggests that SDH deficiency is a contributing mechanism to progressive beta-cell failure in diabetes and identifies mTOR complex 1 inhibition as a potential strategy for mitigation.

Summary: This study examined the associations between COBLL1 rs7607980 and IRS1 rs2943641 gene polymorphisms and the risk of gestational diabetes mellitus (GDM), as well as the expression of these genes in the placenta of GDM women. The results suggest that these gene polymorphisms are not significant risk factors for GDM in the studied population, but the IRS1 TT genotype may be associated with higher fasting glucose levels in GDM women. Additionally, the expression of IRS1 in the placenta positively correlates with an increase in BMI during pregnancy in GDM women.
