Functional evaluation of vandetanib metabolism by CYP3A4 variants and potential drug interactions in vitro

Mingming Han et al.

Summary: This study investigated the enzymatic properties of 27 CYP3A4 variants and their ability to metabolize vandetanib in vitro, finding enhanced and decreased metabolism rates among different variants. Furthermore, the study compared the metabolism of vandetanib and cabozantinib by CYP3A4 variants, as well as the impact of the CYP inhibitor ketoconazole on vandetanib potency in different variants. These findings provide insights into allele-specific metabolism of vandetanib and drug interactions, which can inform the clinical application of vandetanib.

Pharmacokinetic and Pharmacodynamic Considerations in Relation to Calcineurin Usage in Elderly Kidney Transplant Recipients

Amelia R. Cossart et al.

Summary: Age-related changes in physiology can affect drug metabolism and responsiveness, potentially requiring adjustments in drug dosing for elderly patients. Elderly kidney transplant recipients are at higher risk for complications, especially due to increased susceptibility to immunosuppressant side effects; however, current dosing and monitoring protocols may not account for age differences.

Functional assessment of the effects of CYP3A4 variants on acalabrutinib metabolism in vitro

Mingming Han et al.

Summary: Genetic polymorphism extensively impacts CYP3A4's enzymatic activity in metabolizing ACA, showing varied capabilities even within the same variants. The inhibitory potency of CYP inhibitor differs among different variants when combined with ACA. The distance between the substrate and the heme iron atom, likely influenced by mutation, plays a crucial role in determining the enzymatic capabilities of CYP3A4.

Frequency and Risk Factors of Cyclosporine-Induced Neurotoxicity in Allogeneic Stem Cell Transplant Recipients

Asma Danish et al.

Summary: This study investigated the frequency and risk factors of cyclosporine-induced neurotoxicity (CIN) in HLA-matched allogeneic stem cell transplant patients. The results showed that neurological complications, especially seizures, were common in CIN patients, and prompt recognition of symptoms and early intervention can stop the disease progression.
