Impact of adolescent methamphetamine use on social cognition: A human-mice reverse translation study

Antonio Verdejo-Garcia et al.

Summary: The study revealed that individuals with methamphetamine dependence who started using the drug during adolescence exhibited higher antisocial beliefs and poorer emotion recognition compared to those who started using during adulthood. Experiments on mice showed that exposure to methamphetamine during adolescence specifically decreased social investigation during social interaction and had neurochemical effects on the striatal dopaminergic system. These findings suggest heightened sensitivity to methamphetamine effects on social cognition during adolescence.

Mitragynine improves cognitive performance in morphine-withdrawn rats

Chiek Yi You et al.

Summary: The study aimed to examine the cognitive effects of mitragynine substitution in morphine-withdrawn rats and investigate whether neuronal addiction markers such as brain-derived neurotrophic factor (BDNF) and Ca2+/calmodulin-dependent kinase II alpha (alpha CaMKII) might mediate the observed effects. Results showed that mitragynine substitution was able to alleviate morphine withdrawal signs and improve cognitive deficits, suggesting its potential as a safe medication therapy for opiate addiction.

The respiratory depressant effects of mitragynine are limited by its conversion to 7-OH mitragynine

Rob Hill et al.

Summary: The study found differential effects of Mitragynine and its metabolite, 7-OH Mitragynine, on mouse respiration and anti-nociception. Mitragynine showed a ceiling effect on respiratory depression, while 7-OH Mitragynine had dose-dependent effects. Inhibition of CYP3A reduced the respiratory depressant effects and anti-nociception induced by Mitragynine, but had no effect on the effects of 7-OH Mitragynine. These findings suggest that metabolic saturation may contribute to the improved safety profile of Mitragynine.

Microbial-derived metabolites as a risk factor of age-related cognitive decline and dementia

Emily Connell et al.

Summary: The increasing prevalence of age-related cognitive decline and dementia is a consequence of our ageing global population. The microbiota-gut-brain axis, a dynamic bidirectional communication system between the gut, its microbiome, and the central nervous system, may play a role in cognitive health and disease. Microbial-derived metabolites have been identified as potential risk factors for cognitive decline, impacting cognition through cytotoxic metabolite production, neuroinflammation, and other mechanisms.

Activation of proline biosynthesis is critical to maintain glutamate homeostasis during acute methamphetamine exposure

Bobby Jones et al.

Summary: The study reveals the essential role of proline biosynthesis in maintaining METH-induced glutamate homeostasis. METH exposure results in a significant upregulation of pyrroline-5-carboxylate synthase (P5CS), key enzyme for proline synthesis. Depletion of P5CS leads to an increase in glutamate levels upon METH exposure, emphasizing the protective mechanism to maintain glutamate homeostasis.

The Adverse Cardiovascular Effects and Cardiotoxicity of Kratom (Mitragyna speciosa Korth.): A Comprehensive Review

Mohammad Farris Iman Leong Bin Abdullah et al.

Summary: Despite the potential therapeutic value of kratom, studies have found that its most abundant alkaloid, mitragynine, could cause a prolonged QTc interval and an increased risk of torsades de pointes. Regular consumption of kratom may also induce an increased QTc interval in a dose-dependent manner. While kratom use has been associated with ventricular arrhythmia and cardiopulmonary arrest, these events may be due to co-administration with other substances. National poison data analysis indicated that common acute cardiovascular effects of kratom include tachycardia and hypertension. Coroner and autopsy reports highlighted cardiovascular sequelae such as coronary atherosclerosis, myocardial infarction, left ventricular hypertrophy, and cardiac arrhythmia.
