Nanomedicines, an emerging therapeutic regimen for treatment of ischemic cerebral stroke: A review

Wei Chen et al.

Summary: Cerebral stroke, caused by interruption of blood supply to brain tissues, requires prompt risk assessment and therapeutic plan with neuroprotection. Fibrinolytic agents are commonly used but face limitations; nanomedicines show potential in improving drug efficiency for ischemic brain tissues.

Pretreatment of Indobufen and Aspirin and their Combinations with Clopidogrel or Ticagrelor Alleviates Inflammasome Mediated Pyroptosis Via Inhibiting NF-kappa B/NLRP3 Pathway in Ischemic Stroke

Fengyang Li et al.

Summary: The study demonstrates that the combination of indobufen or aspirin with clopidogrel or ticagrelor as pretreatment shows protective effects against cerebral ischemic injury by inhibiting the NF-kappa B/NLRP3 pathway, reducing inflammatory response, and decreasing infarct size, brain edema, LDH release, and neurological impairment. The combination also reduces inflammasome mediated pyroptosis in both in vivo and in vitro models, suggesting a potential clinical strategy for treating ischemia/reperfusion-induced injury.

Challenges and Improvements of Novel Therapies for Ischemic Stroke

Lijun Yang et al.

Summary: Stroke is the third most common disease globally, particularly ischemic stroke causing severe neural cell death. While many therapies show promise in preclinical studies, few pass clinical trials. Novel methods such as traditional Chinese medicine, stem cell therapy, and exosomes are being explored for potential treatment of ischemic stroke.

Molecular pathophysiological mechanisms of ischemia/reperfusion injuries after recanalization therapy for acute ischemic stroke

Anamaria Jurcau et al.

Summary: Recanalization therapy, increasingly used for treating acute ischemic stroke, results in about one-third of patients experiencing early neurological deterioration due to ischemia/reperfusion injuries. Extensive knowledge of the involved mechanisms is crucial for efficient prevention or treatment of these injuries, which include mitochondrial dysfunction, cellular calcium overload, excitotoxicity, oxidative stress, apoptosis, and neuroinflammation. The pathways contributing to the pathophysiology of these injuries have dual effects, being damaging or neuroprotective depending on the timing or protein subtype involved.
