Structural abnormalities of retinal pigment epithelial cells in a light-inducible, rhodopsin mutant mouse

Debora Napoli et al.

Summary: Retinal pigment epithelium (RPE) is a specialized cell layer dedicated to supporting and protecting the retina. In retinitis pigmentosa (RP), degeneration of the photoreceptors can lead to abnormal organization and compromised barrier function of RPE cells. The biological mechanisms behind these changes are largely unknown. A study using a mouse model of RP suggests that inflammatory responses may be involved in remodeling of RPE, and further investigation is needed.

The Potential Role of Epigenetic Mechanisms in the Development of Retinitis Pigmentosa and Related Photoreceptor Dystrophies

Galina Dvoriantchikova et al.

Summary: Retinitis pigmentosa and related photoreceptor dystrophies (RPRPD) are rare retinal diseases caused by hereditary gene mutations. This study suggests that epigenetic changes, specifically DNA methylation and demethylation, may play a significant role in the pathogenesis of these diseases. The research also found that genes required for photoreceptor development and associated with RPRPD have epigenetic restrictions in retinal progenitor cells but not in photoreceptors.

Disease-associated microglial activation prevents photoreceptor degeneration by suppressing the accumulation of cell debris and neutrophils in degenerating rat retinas

Juncai He et al.

Summary: This study investigated the activation of microglia and its contribution to photoreceptor degeneration in retinitis pigmentosa (RP). The results showed that disease-associated microglia activation can suppress the degeneration of rods and cones, reducing secondary damage caused by the accumulation of dead cells and infiltrated neutrophils.

Retinal Inflammation, Cell Death and Inherited Retinal Dystrophies

Lorena Olivares-Gonzalez et al.

Summary: Inherited retinal dystrophies (IRDs) are a group of retinal disorders that lead to progressive vision loss due to retinal cell death, predominantly photoreceptor cells. The treatment of IRDs is complicated by genetic and clinical heterogeneity, but reducing retinal inflammation has emerged as an effective approach to slowing disease progression.

Retinal Pigment Epithelium Remodeling in Mouse Models of Retinitis Pigmentosa

Summary: The study reveals structural abnormalities in the RPE of RP patients, leading to dysfunction of the inner barrier. Findings also suggest a correlation between the extent of photoreceptor loss and interruption zones of ZO-1. Treatment with dexamethasone in RP mice not only rescues cones but also restores RPE structure, highlighting a potential feedback loop for simultaneous protection of cones and the RPE.
