Summary: The dynamics, distribution, and activity of subcellular organelles, particularly mitochondria, play a crucial role in regulating cell shape changes during physiological processes. Recent literature highlights the importance of mitochondrial morphology and activity in various cell shape changes, such as epithelial cell formation, cell division, cell migration, and tissue morphogenesis. The regulation of mitochondrial shape is closely related to their distribution and translocation to sites of active cell shape dynamics. Metabolites released by mitochondria and molecules buffered by mitochondria play critical roles in regulating signaling pathways that drive changes in cell shape, mitochondrial shape, and mitochondrial activity. Understanding the mechanistic interactions between mitochondrial morphology, activity, signaling pathways, and cell shape changes is key to deciphering the common principles underlying this interaction.

100 Hz ROCS microscopy correlated with fluorescence reveals cellular dynamics on different spatiotemporal scales

The authors introduce Rotating Coherent Scattering (ROCS) microscopy, a label-free high contrast imaging method that combines intensity speckle patterns from all illumination directions. With 160 nm resolution and 100 Hz speed, ROCS microscopy allows for imaging thousands of acquisitions in live cells. The authors demonstrate the performance of improved Total Internal Reflection (TIR)-ROCS in combination with fluorescence, and use time-window analysis methods to understand the effects of motion blur on cellular structures and slow structure motions.
