High-Sensitivity Amperometric Dual Immunoassay Using Two Cascade Reactions with Signal Amplification of Redox Cycling in Nanoscale Gap

Summary: In this study, a high-sensitivity dual immunoassay using Lumulus amebocyte lysate (LAL) and blood coagulation cascade reactions with redox cycling in a nanoscale-gap electrode was developed. The dual immunoassay showed high sensitivity for detecting two types of analytes, and the signal amplification by the cascade reactions and the use of redox cycling in a nanoscale gap contributed to the high sensitivity. The cross reaction between the LAL and coagulation cascade reactions was minimized by bisecting the beads after the immunocomplex formation. The dual immunoassay demonstrated its potential for clinical diagnosis requiring the detection of multiple analytes.

Summary: A novel signal-off electrochemical aptasensor was developed for highly sensitive detection of LPS by combining AgNPs-decorated TNT with PDDA-functionalized rGO. The nanohybrid exhibited excellent electrochemical activity, serving as both the sensing platform for LBA immobilization and the redox probe for signal monitoring. The aptasensor showed good stability, reproducibility, specificity, wide linear range, low detection limit, and acceptable recovery rates for real sample analysis.

The method successfully achieved nanomolar sensitivity by utilizing the nanoscale gap for signal amplification.
