Aster repulsion drives short-ranged ordering in the Drosophila syncytial blastoderm

Jorge De-Carvalho et al.

Summary: During the syncytial stage of fruit fly embryo development, nuclei divide synchronously and position themselves with remarkable order. Scientists have discovered that short-ranged repulsive interactions between microtubule asters play a crucial role in ensuring the regular distribution and maintenance of nuclear positions in the embryo.

Experimental and Natural Induction of de novo Centriole Formation

Kasuga Takumi et al.

Summary: Centrioles can be assembled in the vicinity of pre-existing centrioles or in the absence of pre-existing centrioles, a process known as de novo centriole formation. This process can be triggered by the removal of all pre-existing centrioles in the cell or through the overexpression of certain regulatory proteins. De novo centriole formation can occur naturally in some species. This review summarizes previous findings on de novo centriole formation, particularly under experimental conditions, and discusses its regulatory mechanisms.

Female meiosis II and pronuclear fusion require the microtubule transport factor Bicaudal D

Paula Vazquez-Pianzola et al.

Summary: BicD is essential for producing normal female meiosis II products, preventing re-entry of female meiotic products into the cell cycle, and pronuclear fusion. It plays a role in localizing SAC components for metaphase arrest and interacts with Clathrin heavy chain to correctly position proteins during female meiosis II.

Cytological heterogeneity of heterochromatin among 10 sequenced Drosophila species

Marchetti Marcella et al.

Summary: Chromosomal rearrangements in Drosophila contribute to karyotypic variability among populations and provide insights into the evolutionary history of the genus. Comparative analysis of syntenic blocks and physical maps on polytene chromosomes reveals changes in genomic sequences and differences between heterochromatic and euchromatic regions. However, the cytological maps cannot study the centromeric heterochromatic regions. This study expands on the cytological mapping of heterochromatin in 10 Drosophila species, highlighting two different paths of evolution in pericentric heterochromatin between subgenera Sophophora and Drosophila, and provides a better understanding of karyotype alterations due to chromosome arm reshuffling during evolution.

Astral microtubule cross-linking safeguards uniform nuclear distribution in the Drosophila syncytium

Ojas Deshpande et al.

Summary: This study highlights the essential role of Feo and Klp3A in the distribution and maintenance of nuclear internuclear distance in Drosophila embryos through cross-linking of microtubules. Disruption of this cross-linking mechanism leads to irregular nuclear delivery and smaller distribution in the embryos. The dimerization activity of Feo is crucial for maintaining proper nuclear separation and distance in the embryo.

Cytoskeletal mechanics and dynamics in the Drosophila syncytial embryo

Zhiyi Lv et al.

Summary: Cell and tissue functions rely on genetic programmes and biochemical signals, while the physical properties of soft material play an important role in cellular function and morphology. The biophysical properties are determined by the cytoskeleton, consisting of dynamic networks and associated proteins. The Drosophila syncytial embryo, a simple pseudo-tissue, showcases how cytoskeletal dynamics underlies biophysical properties and collective features.

Nuclear positioning during development: Pushing, pulling and flowing

Ojas Deshpande et al.

Summary: Nuclear positioning is a crucial process in cell biology, linked to cell cycle, differentiation, migration, and polarity. While the cytoskeletal elements and force generating elements are well understood, there is still uncertainty about the biophysical mechanism of nuclear positioning, leading to ongoing debate in the field. Further research is needed to explore this area and address open questions.

Parental genome unification is highly error-prone in mammalian embryos

Tommaso Cavazza et al.

Summary: Most human embryos are aneuploid and the origin of aneuploidy during early mitotic divisions remains elusive. Clustering of parental genomes with nucleoli in pronuclei is essential for their reliable unification after fertilization, but failure in clustering often leads to chromosome segregation errors and micronuclei, incompatible with healthy embryo development.

Plk4 triggers autonomous de novo centriole biogenesis and maturation

Catarina Nabais et al.

Summary: Centrioles are formed by centrosomes and cilia. In most proliferating cells, centrioles are assembled through canonical duplication, regulated by the cell cycle and the presence of mature centrioles. However, in certain cell types, centrioles are assembled de novo by mechanisms that are not yet fully understood.

Dual spindles assemble in bovine zygotes despite the presence of paternal centrosomes

Isabell Schneider et al.

Summary: In mammalian embryos, two independent spindles are formed around the genomes of the two pronuclei, despite the presence of centrosomes. Centrosomes have little effect on spindle structure and are loosely connected to the two spindles. This dual spindle assembly pathway is conserved in nonrodent mammals and may explain the frequent loss of whole parental genomes observed in blastomeres of human IVF embryos.
