Impact of immune checkpoint molecules on FoxP3+ Treg cells and related cytokines in patients with acute and chronic brucellosis

Summary: The study found that the frequency of Tregs in chronic brucellosis patients was higher compared to healthy individuals and acute patients. CTLA-4(+) Tregs were significantly higher in chronic patients than in healthy controls, and GITR(+) and PD-1(+) Tregs were significantly higher in acute and chronic patients than in healthy controls. TGF-beta 1 levels were higher in chronic patients, while IL-10 levels were higher in acute patients.

Summary: This study aimed to evaluate the expressions of TFs and cytokines during early infection with different virulent PRRSV-1 strains. The results showed upregulation of TFs T-bet, EOMES, and FOXP3, along with an increase in IFN-gamma, suggesting a stronger polarization to Th1 cells and Tregs in virulent PRRSV-1-infected animals, warranting further studies on their biological functionality.

Up-Regulation of Immune Checkpoints in the Thymus of PRRSV-1-Infected Piglets in a Virulence-Dependent Fashion

Summary: This study aimed to evaluate the expression of selected immune checkpoints in the thymus of piglets infected with two different PRRSV-1 strains. The up-regulation of PD1/PDL1, CTLA4, TIM3, LAG3 and IDO1 immune checkpoints in the thymus of PRRSV-infected piglets, especially those infected with the virulent Lena strain from 6 dpi onwards, was associated with disease progression, high viral load and cell death. Co-expression of these molecules can negatively regulate the host immune response against PRRSV by affecting T-cell development, maturation and selection.

Summary: By comparing the host transcriptional response between PRRSV-infected PAMs and uninfected but exposed bystander PAMs, it was found that inflammatory cytokines, interferon-stimulated genes, and antiviral genes were highly upregulated in infected cells. Additionally, negative immune regulators and T-cell exhaustion markers were also significantly upregulated in infected PAMs compared to bystander cells.

Summary: The study evaluated regulated cell death events by analyzing caspases expression in the lung of PRRSV-1-infected pigs, showing that infection with virulent strains caused severe lung damage and high frequency of infected cells, followed by induction of different forms of cell death in the first week post-infection and mainly activation of extrinsic apoptosis in the second week post-infection.
