The current drug discovery landscape for trypanosomiasis and leishmaniasis: Challenges and strategies to identify drug targets

Fernando Altamura et al.

Summary: Human trypanosomiasis and leishmaniasis, caused by protozoan parasites, have become globally distributed tropical diseases due to human migration, climate change, and anthropogenic disturbance. The current chemotherapy for these diseases has limited efficacy and drug resistance is a growing concern. This review discusses the limitations of available drugs and highlights promising leads from current drug-based clinical trials. It also examines the challenges in target-based drug discovery and the advantages and limitations of modern research tools.

Evaluation of anti-malaria potency of wild and genetically modified Enterobacter cloacae expressing effector proteins in Anopheles stephensi

Hossein Dehghan et al.

Summary: The wild-type and genetically modified strains of Enterobacter cloacae were found to significantly disrupt the development of Plasmodium berghei in the midgut of Anopheles stephensi mosquitoes. This suggests that these bacteria have the potential to provide a barrier or release effector molecules to combat malaria infection.

Comparative analysis of the gut microbiota of sand fly vectors of zoonotic visceral leishmaniasis (ZVL) in Iran; host-environment interplay shapes diversity

Fateh Karimian et al.

Summary: The study analyzed the microbiota within the midguts of different sand fly vectors in Iran. It found highly diverse community composition and abundance, with Proteobacteria being the principal bacterial phylum. Additionally, it found a positive correlation between vector capacity and bacterial diversities, suggesting the potential threat of sand fly vectors to livestock and humans in the region. The study also identified Bacillus subtilis, Asaia sp., and Enterobacter cloacae as possible candidates for reducing Leishmania transmission through a paratransgenic approach.

Cutaneous Leishmaniasis: A 2022 Updated Narrative Review into Diagnosis and Management Developments

Henry J. C. de Vries et al.

Summary: This article is an update of a previous review on the clinical management of cutaneous leishmaniasis (CL), covering diagnosis, treatment, prevention, and control. CL is a global problem, affecting 12 million cases with 2 million new cases annually. Local experience and species determination are crucial for effective management. However, there is a lack of large and well-conducted studies on treatment options, especially for children. Improved vector control, diagnostics, and safe vaccines are urgently needed, but limited investment from biotechnical companies hinders progress.

The optimization of PpSP15 purification from salivary glands in Iranian wild Phlebotomus papatasi (Diptera: Psychodidae)

S. Ghafari et al.

Summary: In this study, a new saliva protein PpSP15 was discovered from sand fly specimens in Iran, and a more effective chemical method for protein extraction was developed. This is important for developing a vaccine against leishmaniasis, as suitable proteins should be chosen based on location and an efficient extraction method should be used.

Immune Responses in Leishmaniasis: An Overview

Ana Caroline Costa-da-Silva et al.

Summary: Leishmaniasis is a widespread and neglected parasitic disease caused by various species of Leishmania, with severity depending on multiple factors. This article discusses the role of the immune system, its molecules, and responses in the development of Leishmaniasis, focusing on the interactions between innate immune cells and Leishmania major.

Role of host genetics and cytokines in Leishmania infection

Imtissal Krayem et al.

Summary: The study highlights the impact of host genetic background, parasite species and sub-strain on the effects of genetically engineered cytokine and chemokine genes in leishmaniasis. It also reveals a complex network-like interaction of multiple functionally diverse QTLs and their individual specificity, with only partial overlap between genetic control of organ pathology and systemic immune response.

Enterobacter hormaechei in the intestines of housefly larvae promotes host growth by inhibiting harmful intestinal bacteria

Qian Zhang et al.

Summary: This study found that adding Enterobacter hormaechei can change the gut microbial community of housefly larvae, promote the reproduction of beneficial bacteria, and inhibit the growth of harmful bacteria, providing new possibilities for housefly control.

Prevalence of Genetically Complex Leishmania Strains With Hybrid and Mito-Nuclear Discordance

Hirotomo Kato et al.

Summary: The discovery of unknown hybrid Leishmania parasites in Ecuador and Peru using PCR-RFLP targeting nDNA, alongside species reported previously, suggests that genetic exchange may occur more frequently in natural populations than previously thought. Comparative analyses of kDNA and nDNA revealed mismatches between genes, representing the first report of mito-nuclear discordance in protozoa, indicating the need for further exploration into how such genetic exchange influences disease progression and transmissibility by sand flies in nature.

Leishmaniases in the European Union and Neighboring Countries

Eduardo Berriatua et al.

Summary: A questionnaire survey in Europe found that leishmaniases are not universally notifiable in countries with autochthonous cases. Few countries have surveillance and control measures in place for both animal and human infections. Lack of resources is a major challenge for controlling leishmaniases, which are considered emergent diseases in most countries.
