Ribonucleotide reductase, a novel drug target for gonorrhea

Jana Narasimhan et al.

Summary: Novel inhibitors PTC-847 and PTC-672 were found to exhibit a narrow spectrum of activity against multidrug-resistant Neisseria gonorrhoeae (Ng). The study identified a new therapeutic target, class Ia ribonucleotide reductase (RNR), and revealed the involvement of the N-terminal cone domain of the alpha subunit in resistance mutations. Oral administration of PTC-672 showed potential in reducing Ng infection in a mouse model.

Summary: S100 is a broad subfamily of low-molecular weight calcium-binding proteins with structural similarity and functional discrepancy. It plays a role in inflammation, cellular homeostasis, and has both extracellular and intracellular functions. Several members of the S100 family are implicated in the pathophysiology of COVID-19. S100 proteins may serve as prognostic markers for COVID-19 infection identification.

Self-sacrificial tyrosine cleavage by an Fe:Mn oxygenase for the biosynthesis of para-aminobenzoate in Chlamydia trachomatis

Olivia M. Manley et al.

Summary: Chlamydia protein associating with death domains (CADD) is involved in the biosynthesis of para-aminobenzoate (pABA) from L-tyrosine in Chlamydia trachomatis. The addition of both iron and manganese leads to the maximal formation of pABA by CADD, suggesting a heterobimetallic Fe:Mn cluster as the catalytically active form. The reaction mechanism involves O-2 activation by the Fe:Mn cluster, followed by amination and direct oxygen insertion. These findings provide insights into pABA formation in C. trachomatis.

Redox-controlled reorganization and flavin strain within the ribonucleotide reductase R2b-NrdI complex monitored by serial femtosecond crystallography

Juliane John et al.

Summary: Redox reactions play a central role in biochemistry and are controlled by protein structural changes. In this study, the authors utilized femtosecond crystallography to examine the structural rearrangements and crosstalk within the Bacillus cereus ribonucleotide reductase R2b-NrdI complex. They found that the flavin in the complex is under steric strain, which likely affects its redox properties and promotes superoxide generation. The redox state of the flavin also controls a binding site that is linked to the channel responsible for transporting superoxide species. These findings have important implications for understanding the control of reactive oxygen species and radical generation in protein R2b.

Jan Capek et al.

Summary: The human-restricted pathogen Bordetella pertussis has evolved a unique reverse adaptation mechanism through an inhibitory duplication within a manganese exporter gene, allowing it to accumulate manganese and improve survival under stress conditions. This specialized process assists B. pertussis in adjusting to environments with varying levels of manganese, enhancing its resistance to both toxicity and scarcity of the nutrient.

Inter-domain horizontal gene transfer of nickel-binding superoxide dismutase

K. M. Sutherland et al.

Summary: The regulatory role of SOD enzymes, especially SodN, in aerobic microorganisms is crucial for cell health and survival. Evolutionary history shows convergence in the genes encoding different SOD enzymes, driven by environmental selection factors like oxygen levels and metal availability. Horizontal gene transfer plays a significant role in the diversity and distribution of SodN, with evidence of multiple transfers between bacterial and archaeal domains. Additionally, the widespread presence of sodN in various microbial groups suggests its adaptation to changing environmental conditions, particularly in response to decreasing iron availability in the surface ocean.

Analysis of the Manganese and MntR Regulon in Corynebacterium diphtheriae

Eric D. Peng et al.

Summary: Corynebacterium diphtheriae is the causative agent of severe respiratory disease, with metal acquisition, particularly manganese, critical for colonization. The Mn-dependent regulator MntR in C. diphtheriae modulates the expression of various target genes, including Mn-superoxide dismutase and putative ABC transporter. DNA binding studies identified a consensus MntR DNA binding site, with evidence suggesting the MntABCD transporter functions as an Mn uptake system. This study enhances understanding of metal regulatory networks in this pathogen and may aid in vaccine development.

Yersiniabactin contributes to overcoming zinc restriction during Yersinia pestis infection of mammalian and insect hosts

Sarah L. Price et al.

Summary: Research has shown that the iron-scavenging siderophore Ybt produced by Yersinia pestis not only helps overcome iron-mediated nutritional immunity, but may also contribute to overcoming zinc-mediated nutritional immunity. Furthermore, it was discovered that Ybt competes with calprotectin for zinc during Y. pestis infection, aiding the bacteria in overcoming zinc limitation. Additionally, Ybt assists in surmounting zinc limitation within the flea midgut, serving as a bona fide zinc acquisition mechanism for Y. pestis during infection of both mammalian and insect hosts.
